摘  要:目的:探讨血清miRNA-4516在肝癌中的表达情况及其临床意义并预测其可能的靶基因。方法:收集100例肝癌患者和同期体检的100例健康人血清样本,采用RT-PCR测定血清miRNA-4516表达水平,分析其与肝癌临床病理的相关性,最后通过生物信息学预测miRNA-4516可能的靶基因。结果:经分析血清miRNA-4516相对表达水平,肝癌患者高于正常对照组(P=0.001);与肝癌的分化程度有关(P=0.015);而与患者年龄、性别、AFP表达量、是否肝炎肝硬化、是否转移无相关性(P>0.05)。通过生物信息学分析,miRNA-4516预测的靶基因多数与转录因子有关。结论:miRNA-4516有可能通过调节转录因子在肝癌发生、发展中起作用。Objective: This study sought to detect the expression and clinical significance of serum miRNA-4516 in hepatocellular carcinoma, and to predict its target genes through bioinformation. Methods: A total of 100 patients with hepatocellular carcinoma and 100 healthy persons were selected. The level of serum miRNA-4516 was detected by RT-PCR. The correlation of serum miRNA-4516 expression levels with the clinical characteristics of hepatocellular carcinoma was analyzed. Bioinformatics methods were used to analyze target genes of miRNA-4516. Results: Serum miRNA-4516 was highly expressed in patients with hepatocellular carcinoma in contrast with controls(P=0.001). The high expression of miRNA-4516 was shown to be related to differential grade of hepatocellular carcinoma(P=0.015), Whereas the high expression of miRNA-4516 didn’t correlate with age, sex, AFP, hepaitis, cirrhosis and distant metastasis of the patients(P>0.05). Target genes predictived by information are largely transcription factor. Conclusion: miRNA-4516 may play a role in the occurrence and development of hepatocellular carcinoma through transcription factor.
