摘  要:目的探讨芪归健脑方对阿尔茨海默病(AD)大鼠神经组织中的基质金属蛋白酶-9(MMP-9)、闭合蛋白-5(claudin-5)和咬合蛋白(occludin)含量的影响。方法制备AD大鼠模型,造模后进行不同的处理。芪归组:每天口服芪归健脑方1 g/kg,吡拉西坦组:每天口服吡拉西坦0.43 g/kg;对照组与AD组:口服同样剂量的无菌生理盐水,疗程均为30天。所有实验对象投入水迷宫检测其行为变化;免疫印迹评估大脑组织中MMP-9、claudin-5和occludin的含量的改变。结果与对照组比较,AD组大鼠的学习记忆功能明显减弱,MMP-9的水平(0.572±0.031)上升,而claudin-5(0.188±0.021)和occludin(0.192±0.021)的水平降低(均P<0.05);口服芪归健脑方后其学习记忆功能明显改善,MMP-9的水平(0.338±0.017)下降,而claudin-5(0.295±0.014)和occludin(0.296±0.011)的水平升高(均P<0.05)。芪归组与吡拉西坦组比较,差异无统计学意义(P>0.05)。结论芪归健脑方可以改善AD大鼠大脑的学习记忆功能,这与其能下调MMP-9的水平,上调claudin-5和occludin水平紧密相关。Objective To explore the effect of Qigui Jiannao Prescription on expression of matrix metalloproteinases-9(MMP-9),claudin-5 and occludin in hippocampus of rats with Alzheimer’s disease(AD).Methods AD rat models were prepared and then treated differently.Qigui group was given Qigui Jiannao Prescription 1 g/kg per day orally,piracetam group was given piracetam 0.43 g/kg per day orally,while control group and AD group were given equivalent of normal saline orally,and the course of treatment was 30 days.All subjects were put into a water maze to detect changes in their behavior.Western blots were used to assess content changes in MMP-9,claudin-5,and occludin in brain tissues.Results Compared with control group,the learning and memory function of AD group was obviously decreased,the level of MMP-9(0.572±0.031)was increased,and the levels of claudin-5(0.188±0.021)and occludin(0.192±0.021)were decreased(all P<0.05).After oral administration of Qigui Jiannao Prescription,the learning and memory functions were obviously improved,the level of MMP-9(0.338±0.017)was decreased,while the levels of claudin-5(0.295±0.014)and occludin(0.296±0.011)were increased(all P<0.05).The difference was not statistically significant between Qigui group and piracetam group(P>0.05).Conclusion Qigui Jiannao Prescription can improve the learning and memory function of AD rat,which is closely related to the decrease of MMP-9 level and the increase of claudin-5 and occludin levels.
