摘  要:以淡水鱼内脏为原料,以鱼油得率为评价指标,对酶法提取鱼油的工艺条件进行优化。首先采用木瓜蛋白酶,中性蛋白酶和碱性蛋白酶在各自的最佳酶活条件下提取内脏鱼油,得到最佳的水解用酶为中性蛋白酶。然后以中性蛋白酶为水解用酶,在单因素试验基础上,采用正交试验设计,分别考察pH值、中性蛋白酶添加量、酶解温度、酶解时间和料液比对鱼油得率的影响,最终得到中性蛋白酶酶解法提取鱼油的最优条件为:pH值7,酶解时间40 min,酶解温度60℃,酶添加量0.50%,料液比(原料∶蒸馏水)1∶1.5。在最优条件下,鱼油得率可达35.67%。With fresh water fish offal as the raw material and fish oil yield as the evaluation index,the extraction conditions of fish oil by enzymatic method are optimized.Firstly,papain,neutral protease and alkaline protease are used to extract offal fish oil under their respective optimal enzyme activity conditions,and the optimal hydrolysis enzyme is neutral protease.Secondly,the neutral protease is used as the hydrolysis enzyme,the orthogonal experiment design is adopted to investigate the effects of pH value,additive amount of neutral protease,enzymatic hydrolysis temperature,enzymatic hydrolysis time and solid-liquid ratio on the yield of fish oil based on the single factor experiment.Ultimately,the optimal conditions for extracting fish oil by neutral protease hydrolysis are as follows:pH value is 7,enzymatic hydrolysis time is 40 min,enzymatic hydrolysis temperature is 60℃,enzyme additive amount is 0.50%,the ratio of solid to liquid(raw material∶distilled water)is 1∶1.5.Under the optimal conditions,the yield of fish oil can reach 35.67%.
