摘  要:目的:观察γ-谷酰基转移酶(gamma-glutamyl transferase,GGT)在小鼠皮肤成纤维细胞中的表达,探究抑制其活性后对细胞存活率及Ⅰ型胶原表达的影响。方法:胰酶、胶原酶两步消化法分离新生C57BL/6小鼠皮肤成纤维细胞并在培养板中培养至稳定贴壁生长;免疫组化方法检测GGT在小鼠皮肤成纤维细胞中的表达;生长噻唑蓝(MTT)法检测不同浓度GGsTOP对体外培养小鼠皮肤成纤维细胞存活率的影响;免疫荧光观察COL-Ⅰ的表达。结果:GGT在小鼠皮肤成纤维细胞中广泛表达。抑制剂GGsTOP可以促进小鼠皮肤成纤维细胞增殖,并且使细胞内Ⅰ型-胶原(type Ⅰ collagen,COL-Ⅰ)蛋白水平的表达增强。结论:GGT在小鼠皮肤成纤维细胞中广泛表达,抑制GGT酶活性后促进Ⅰ型-胶原表达,提示抑制剂GGsTOP可能促进创面愈合再上皮化过程。Objective:To observe the gamma glutamyl transferase(GGT)expression on mouse skin fibroblasts(MSF),and explore whether inhibitory activity of GGT influence cell survival and collagen typeⅠ(COL-1)expression.Methods:Primary mouse dermal fibroblast cells were isolated from neonatal C57BL/6 mice by trypsin-collagen two-step digestion.The expression of GGT was analyzed by immunohistochemistry,and the effects of GGsTOP on proliferation of mouse skin fibroblasts cells were evaluated by MTT assay.The collagen typeⅠexpression wasmeasured by immunofluorescence.Results:GGT was widely expressed on mouse skin fibroblasts.Inhibitors GGsTOP promoted mouse skin fibroblasts proliferation,and immunofluorescence showed that the protein expressions of COL-1were up-regulated.Conclusion:GGT was widely expressed in mouse skin fibroblast cells,inhibitors GGsTOP was related to the up-regulation of the expression of COL-1 while inhibition activity of GGT,inhibitors GGsTOP may promote on the re-epithelialization process of wound healing.
