摘  要:目的:探究转录因子SNAI2(zinc-finger transcription factor-2)在口腔鳞状细胞癌中的表达及其对细胞增殖侵袭能力的影响。方法:临床收集53例口腔鳞状细胞癌样本,反转录-聚合酶链反应(RT-PCR)及免疫组化(IHC)检测样本中SNAI2 mRNA及蛋白表达,方差分析及χ2检验比较组间水平差异。慢病毒转染SNAI2干扰表达载体,稳定干扰人口腔鳞癌细胞SCC-090 SNAI2的表达,RT-PCR及Western Blot进行mRNA及蛋白水平的验证,噻唑蓝比色法(MTT)检测细胞增殖能力的变化,transwell侵袭实验检测细胞侵袭能力的变化,Western Blot检测侵袭相关因子金属基质蛋白酶2(MMP-2)及金属基质蛋白酶9(MMP-9)的表达变化。结果:口腔鳞状细胞癌组织中SNAI2 mRNA及蛋白的表达均显著高于癌旁组织,差异有统计学意义(P<0.05)。稳定干扰SNAI2表达后的SCC-090细胞系(SCC-090 shSNAI2)SNAI2 mRNA及蛋白表达水平均显著下降。SCC-090 shSNAI2细胞增殖能力在48及72h时出现有统计学意义的降低(P<0.05),transwell侵袭实验示SCC-090 shSNAI2细胞侵袭能力也显著降低(P<0.05)。SCC-090 shSNAI2的MMP-2及MMP-9蛋白表达相比对照组也显著下降。结论:人口腔鳞状细胞癌中SNAI2高表达,其能促进癌细胞的增殖能力,且能通过调节MMP的表达促进细胞侵袭能力,可能成为新的抗口腔鳞状细胞癌的分子靶点。Objective:To investigate the expression of SNAI2(zinc-finger transcription factor-2)in oral squamous cell carcinoma and its effect on cell proliferation and invasion.Methods:Clinical samples from 53 cases of oral squamous cell carcinoma(OSCC)samples were collected,and the expression level of SNAI2 mRNA and protein was detected by reverse transcription polymerase chain reaction(RT-PCR)and Immunohistochemistry(IHC).The difference between the expression level of SNAI2 mRNA and positive rate of several groups was compared by variance analysis and Chi square test.Lentivirus transfection was used to stably interfere the expression of SNAI2 in SCC-090,and RT-PCR and Western Blot was used to verified in mRNA and protein levels.MTT colorimetric assay(MTT)was used to detect the changes of cell proliferation ability,Transwell invasion assay was used to detect the changes of cell invasion ability,Western Blot was used to detect the expression of metal metalloproteinase 2(MMP-2)and matrix metalloproteinase 9(MMP-9)in shSNAI2 cell line.Results:The expression of SNAI2 mRNA and protein in oral squamous cell carcinoma was significantly higher than that in adjacent tissues,and the difference was statistically significant(P<0.05).The expression of SNAI2 mRNA and protein in SCC-090 cell line(SCC-090 shSNAI2)were significantly decreased after SNAI2 interference.The proliferation of SCC-090 shSNAI2 cells was statistically significant decreased(P<0.05)at 48 and 72h(shSNAI2),and the invasion of Transwell cells showed that the invasion ability of SCC-090 cells was significantly lower(P<0.05).The expression of MMP-2 and MMP-9 in SCC-090 shSNAI2 was significantly lower than that in control group.Conclusion:SNAI2 high expression in human luminal squamous cell carcinoma can promote the proliferation of cancer cells and promote cell invasion by regulating the expression of MMP,which may become a new molecular target against oral squamous cell carcinoma.
