摘 要:低氧是恶性肿瘤微环境的特征之一,缺氧诱导因子-1(HIF-1)是细胞适应低氧环境的重要调节因子。HIF-1是低氧环境下的氧调节型α亚基与组成型β亚基形成的稳定的异二聚体,其促进肿瘤血管生成、上调并激活基质金属蛋白酶及赖氨酸氧化酶等细胞因子及信号通路、诱导特异性微RNA生成,参与多种恶性肿瘤细胞的增殖、侵袭、转移等相关生物学行为。通过不同的方式抑制HIF-1α的活性,改变其结构稳定性,对于恶性肿瘤的治疗具有重要意义。但HIF-1α能否成为恶性肿瘤治疗的靶点及其预后判断的指标,有待于进一步的研究探索。Hypoxia is one of the characteristics of the malignant tumor microenvironment.Hypoxia-inducible factor-1(HIF-1)is an important regulator of cells adapting to the hypoxic environment.HIF-1 is a stable heterodimer formed by oxygen-regulatedαsubunit with constitutiveβsubunit under hypoxic environment.It promotes tumor angiogenesis,upregulates and activates matrix metalloproteinases and lysine oxidase and other cytokines and signal pathways,induces specific miRNA production,and participates in the biological behaviors of proliferation,invasion and metastasis of various malignant tumor cells.Inhibiting the activity of HIF-1αin different ways and changing its structural stability are of great significance for the treatment of malignant tumors.However,whether HIF-1αcan become a target of malignant tumor treatment and its prognostic index needs to be further studied and explored.
