摘 要:目的旨在研究由印迹基因GRB10转录而来的环状RNA hsacirc0008334的基本特征和功能预测。方法通过PCR扩增和一代测序、RNase R处理以及细胞质细胞核分布来研究hsacirc0008334的基本特征。此外,利用生物信息学方法预测hsacirc0008334上结合的miRNA,并进而预测miRNA的靶基因,构建circRNA-miRNA-mRNA网络,利用STRING数据库构建靶基因蛋白的交互作用网络,并对靶基因进行功能富集分析。结果hsacirc0008334是由GRB10基因第5号外显子反向剪接至第4号外显子形成,抵抗RNase R处理,主要分布于细胞质中。生信预测hsacirc0008334通过作为hsa-miR-1265、hsa-miR-187-3p、hsa-miR-586、hsa-miR-604、hsa-miR-941五个miRNA的海绵来发挥作用;五个miRNA的靶基因在对凋亡、细胞程序性死亡、代谢过程等的正向调控和负向调控的生物过程显著富集。结论成功分析了GRB10基因的新型环状RNA hsacirc0008334的基本特征,从circRNA-miRNA-mRNA角度为hsacirc0008334的功能研究提供了新的见解,扩充了GRB10基因的相关研究。Objective To study the feature and function prediction of circular RNA hsacirc0008334 transcribed from imprinted gene GRB10.Methods We have investigated the feature of hsacirc0008334 through PCR amplification,sanger sequencing,RNase R treatment,and the cellular distribution.Additionally,bioinformatic methods were used to predict the miRNA binding sites and the miRNA target genes of hsacirc0008334.A circRNA-miRNA-mRNA regulatory network was constructed and a protein-protein interactions network of target genes was also constructed.Moreover,Gene Oncology(GO)analysis on the miRNA target genes were performed.Results Hsa circ 0008334 was generated through backsplicing of exon 5 to exon 4 of GRB10 gene;hsacirc0008334 was resistant to RNase R treatment and was predominantly localized in the cytoplasm.Hsacirc0008334 may function as sponge of hsa-miR-1265,hsa-miR-187-3 p,hsa-miR-586,hsa-miR-604 and hsa-miR-941.Target genes of the five miRNAs were significantly enriched in multiple biological processes of GO,such as regulation(or negative regulation)of apoptosis,programmed cell death and metabolic process.Conclusion Our study successfully characterizes the novel circRNA hsacirc0008334 derived from GRB10 gene,and provides a novel insight into functional research of hsacirc0008334 from circRNA-miRNA-mRNA network view.Moreover,our study further expands the study of GRB10 gene.
