摘 要:目的:探讨大蒜活性成分二烯丙基三硫化物(diallyl trisulfide,DATS)能否干预香烟烟雾促进的SGC-7901源胃癌干细胞干性,并探究Wnt通路在其中的作用。方法:用不同浓度(0%、0.25%和0.50%)的香烟烟雾悬液和DATS单独或联合处理SGC-7901源胃癌干细胞7 d,显微镜下观察胃癌干细胞球大小及数量的变化,实时定量PCR及蛋白质印迹检测干性基因NANOG、OCT-4、SOX2、LIN28的mRNA和蛋白表达;香烟烟雾悬液、DATS和氯化锂单独或联合处理胃癌干细胞,蛋白质印迹检测干性基因及Wnt通路相关蛋白表达的变化。结果:DATS能明显抑制香烟烟雾促进的胃癌干细胞球的形成及干性基因NANOG、OCT-4、SOX2、LIN28的mRNA和蛋白表达;蛋白质印迹结果显示,DATS能抑制香烟烟雾诱导的β-连环蛋白、糖原合成酶激酶3β(GSK3β)表达异常;氯化锂能增强NANOG和OCT-4的表达,与对照组相比,DATS和氯化锂联合处理组GSK3β蛋白表达水平下调,而β-连环蛋白表达水平上调,同时NANOG和OCT-4的蛋白表达水平上调。结论:DATS能抑制香烟烟雾促进的SGC-7901源胃癌干细胞干性,Wnt通路在其中发挥重要作用。Objective:To investigate whether diallyl trisulfide(DATS)reverse the stemness of SGC-7901-derived gastric cancer stem cells promoted by cigarette smoke,and explore the role of Wnt pathway.Methods:The gastric cancer stem cells obtained from SGC-7901 cells were treated with different concentrations of cigarette smoke extract(CSE,0,0.25%and 0.50%)and DATS alone or in combination for 7 days.The size and number of gastric cancer stem cell spheres under microscope were observed.Real-time quantitative PCR and Western blotting were used to detect the expression of stemness genes NANOG,OCT-4,SOX2 and LIN28.CSE,DATS and Licl were used alone or in combination to treat gastric cancer stem cells.Western blotting were used to detect the expression of stemness genes and Wnt pathway.Results:Compared with CSE alone groups,DATS significantly inhibited the formation of gastric cancer stem cell spheres promoted by CSE and the expression of NANOG,OCT-4,SOX2 and LIN28;Western blotting results showed the abnormal expression ofβ-catenin and GSK3βprotein levels caused by CSE were inhibited.LiCl could enhance the expression of stemness genes NANOG and OCT-4.Compared with the control group,the expression of GSK3βprotein level in DATS and LiCl group was down-regulated,while the expression ofβ-catenin protein level was up-regulated;the expression of NANOG and OCT-4 protein levels were up-regulated.Conclusion:DATS inhibited the stemness of SGC-7901-derived gastric cancer stem cells promoted by cigarette smoke,which may attribute to regulation of the Wnt pathway.
