摘  要:洛蒙真菌素是由洛蒙德链霉菌合成的具有广谱抗菌活性的吩嗪类化合物。luxR调控基因是细菌中一类非常重要的调控基因,广泛参与调控细菌的初级代谢和次级代谢过程。本研究分析了洛蒙德链霉菌S015中两个luxR基因luxR4和luxR13对洛蒙真菌素合成的影响。通过在野生株中单基因过表达luxR4与luxR13,发现这两个基因均能促进洛蒙真菌素的合成,菌株S015L4和S015L13中洛蒙真菌素的产量分别达到野生株的3.7倍和2.4倍。进一步通过luxR4和luxR13双基因共表达,得到高产洛蒙真菌素的菌株S015L4-13,其洛蒙真菌素的产量达到野生株的4.2倍,为(230.2±8.3)mg/L。结果表明,基因luxR4和luxR13对洛蒙真菌素的合成起正调控作用,且两个基因具有一定的协同作用。本研究为揭示洛蒙真菌素的合成的调控机制及高产菌株的构建提供了依据。Lomofungin is a phenazine antibiotics with broad spectrum antibacterial activity,biosynthesized by Streptomyces lomondensis.LuxR are essential regulatory gens in bacteria which involve in the regulation of primary metabolism and secondary metabolism.In this study,the effects of luxR4 and luxR13,in Streptomyces lomondensis S015 on the synthesis of lomofungin were studied through gene overexpression.It was found that both genes luxR4 and luxR13 could promote the synthesis of lomofungin,by overexpressing respectively in wild type and the production of lomofungin in strain S015 L4 and S015 L13 reached 3.7 and 2.4 times respectively of the wild type.Furthermore,by co-expression of luxR4 and luxR13 double genes,a high-yielding lomofungin strain S015 L4-13 was obtained with lomofungin production of(230.2±8.3 mg/L),reached 4.2 times of the wild type.It indicates that genes luxR4 and luxR13 are positive regulators of lomofungin synthesis and cell growth,furthermore,they are synergism.This research laid a foundation for further study on the regulatory mechanism of lomofungin biosynthesis and the construction of high-yield strains.
