摘 要:喀斯特地貌是一种独特的以碳酸盐岩石为主要成分的地理地貌,云南喀斯特地貌就属其中的代表地区之一。吲哚乙酸(indole-3-acetic acid,IAA)是一种常见的植物生长和发育调控激素。IAA产生微生物对植物生长具有促进作用,在农业和植物生长修复中具有广阔应用前景。本研究分析了采自云南东南部5个典型喀斯特地区的102份土样,并对土样中的可培养细菌进行分离和鉴定。共分离、纯化保存795株不同细菌菌株。根据16S rDNA序列分析,这些菌株属于23个属,99个不同种,属于4个门级分类单元:厚壁菌门(Firmicutes)占83.65%;变形菌门(Proteobacteria)占13.21%;放线菌门(Actinobacteria)占2.89%;拟杆菌门(Bacteroidetes)占0.25%。从这些菌株中鉴定出19个不同属的能够产生IAA的细菌56种,共计429个菌株(53.96%),其中产IAA的优势菌群为芽孢杆菌(Bacillus spp.),约占IAA产生菌株的85.24%。结合与土壤因子的关联分析,本研究讨论了IAA产生菌的分子演化趋势并提出了下一步需要深入研究的问题。本研究结果的意义在于进一步丰富了中国喀斯特地貌土壤中IAA产生细菌的种群名录,为进一步挖掘整理、开发并利用这一有益菌群奠定了良好的资源基础。Karst landform is a unique geographical landform mainly composed of carbonate rocks.Karst landform in Yunnan province is one of the representative regions.Indole-3-acetic acid(IAA)is a common hormone regulating plant growth and development.The IAA-producing microbes can promote the growth of plants and have a broad application prospect in agriculture and plant growth remediation.This study analyzed the environmental factors of 102 soil samples from five typical karst areas in southeastern Yunnan,and isolated and identified the culturable bacteria in the soil samples.A total of 795 different bacterial strains were isolated,purified and preserved.According to the 16S rDNA sequence analysis,these strains belonged to 23 genera and 99 different species respectively,belonging to 4 phylum taxa:Firmicutes,accounting for 83.65%;Proteobacteria,accounting for 13.21%;Actinobacteria,accounting for 2.89%;and Bacteroidetes,accounting for 0.25%.Fifty-six species of IAA-producing bacteria belonging to 19 different genera were identified from these strains,with a total of 429 strains(53.96%),of which the dominant IAA-producing bacteria were Bacillus spp.,accounting for about 85.24%of the IAA-producing strains.Taken with the correlation analysis of soil factors,this study also discussed the molecular evolution trend of IAA-producing bacteria and proposed the issues that need further study.The significance of this study lies in that it further enriches the population list of IAA-producing bacteria in the soil of karst topography in China,and lays a good resource foundation for further excavation,sorting,developing and utilizing these beneficial bacteria.
