摘 要:本研究旨在于研究体外华蟾素对人胃癌MGC-803细胞增殖及细胞周期的影响,探究其可能的分子机制。通过台盼蓝拒染法检测华蟾素对人胃癌MGC-803细胞的增殖抑制作用利用激光共聚焦显微镜观察华蟾素作用人胃癌MGC-803细胞48 h后细胞形态,流式细胞术检测细胞周期,实时荧光定量PCR和Western blotting检测CCNB1和CCND1基因的表达情况。结果显示,华蟾素作用人胃癌MGC-803细胞24 h、48 h和72 h的IC50值分别为118.336μg/L、74.063μg/L和49.560μg/L,随着华蟾素浓度的增加,显微镜下细胞出现凋亡特征,MGC-803细胞G1期细胞所占比例逐渐降低,S期细胞数量逐渐增多;CCNB1和CCND1基因表达量降低。研究结果表明,华蟾素能使人胃癌MGC-803细胞出现凋亡特征细胞被阻滞于S期,细胞的增殖分裂被抑制,CCNB1和CCND1基因表达量降低。To investigate effect of Cinobufotalin on the prolifertion and cell cycle of human gastric cancer MGC-803 cells in vitro,and analyze its molecular mechanisms.The trypan blue exclusion was adopted to detect the effect of Cinobufotalin on cell proliferation,the changes of cell morphology was observed under laser scanning confocal microscopy,the change of cell cycle were detected by FACS,and the expression of CCNB1,CCND1 gene mRNA and protein were tested by qRT-PCR and Western blotting,respectively.The results showed that in a certain concentration range,the proliferation of human gastric cancer MGC-803 cells can be inhibited in a dose-dependent manner.The IC50 values of gastric cancer MGC-803 cells were 118.336μg/L,74.063μg/L,49.560μg/L for 24 h,48 h,72 h.With the increase of the concentration of Cinobufotalin,the number of cells decreased under microscope;G1 phase cell reduced,S phase cells increased;The expression level of CCNB1 and CCND1 genes decreased significantly.Conclusion:Cinobufotalin can inhibit the proliferation of human gastric cancer MGC-803 cells,block the cells in S phase,and decrease the expression of CCNB1 and CCND1 genes.
