摘　　要：目的检测胃癌患者血清和胃癌组织中结肠癌转移相关基因1(MACC1)的表达水平,分析血清及胃癌组织中MACC1表达的相关性,并对MACC1作为胃癌发病标记物诊断的有效性进行探讨。方法收集51例胃癌患者的血清标本、胃癌组织、癌旁组织、正常胃黏膜标本,同时收集30例正常人血清标本。分别通过和免疫组织化学法和酶联免疫吸附法(ELISA)测定MACC1在胃癌组织、癌旁组织、正常胃黏膜及血清中的表达水平。分析MACC1在血清与胃癌组织中表达的关系,并绘制受试者操作特征(ROC)曲线。分析MACC1的表达与临床病理参数(性别、年龄、发生部位、肿瘤大小、病理分型、胃壁浸润深度、局部淋巴结转移、远处转移、TNM分期)的关系。结果与健康者相比,胃癌患者血清MACC1表达显著升高(P<0.05)。与胃黏膜组织、癌旁组织相比,胃癌组织中MACC1的表达显著升高(均P<0.05),癌旁组织和正常组织中MACC1的表达差异无统计学意义(P>0.05)。胃癌组织和胃癌患者血清中的MACC1水平呈正相关(P<0.01)。胃癌患者血清及胃癌组织中MACC1水平与患者临床病理参数间均无统计学意义(均P>0.05)。结论MACC1在胃癌患者血清及胃癌组织中均高表达,且表达水平呈正相关。MACC1有望成为诊断胃癌的临床生物指标。Objective To detect the expression level of metastasis-associated in colon cancer 1(MACC1)in the serum and gastric cancer tissue of patients with gastric cancer,analyze the correlation between the expression of MACC1 in the serum and gastric cancer tissue,and evaluate the effectiveness of MACC1 as a marker for the diagnosis of gastric cancer.Methods Serum samples,gastric cancer tissues,adjacent tissues and normal gastric mucosa samples from 51 patients with gastric cancer were collected.At the same time,30 normal human serum samples were collected.The expression levels of MACC1 in gastric cancer tissues,adjacent tissues,normal gastric mucosa and serum were determined by immunohistochemistry and enzyme-linked immunosorbent assay(ELISA).The relationship between MACC1 expression in serum and gastric cancer tissue was analyzed,and the receiver operating characteristic(ROC)curve was drawn.The relationship between the expression of MACC1 and clinicopathological parameters(gender,age,site of occurrence,tumor size,pathological type,depth of gastric wall invasion,local lymph node metastasis,distant metastasis,TNM stage)were analyzed.Results Compared with healthy people,the expression of serum MACC1 in patients with gastric cancer was significantly increased(P<0.05).Compared with gastric mucosal tissues and adjacent tissues,the expressions of MACC1 in gastric cancer tissues were significantly increased(all P<0.05),and there was no statistically significant difference in the expression of MACC1 in adjacent tissues and normal tissues(P>0.05).The level of MACC1 in gastric cancer tissue and serum of gastric cancer patients was positively correlated(P<0.01).There were no statistical significances between the level of MACC1 in serum and tissue of gastric cancer and the clinical pathological parameters of patients(all P>0.05).Conclusions MACC1 is highly expressed in the serum and tissues of gastric cancer patients,and the expression level is positively correlated.MACC1 is expected to become a clinical biomarker for the diagnosis
