摘 要:目的:探讨二氢杨梅素(dihydromyricetin,DMY)增强肺癌A549细胞对顺铂化疗敏感性的作用及机制,为肺癌减毒增敏治疗提供新思路。方法:体外培养A549细胞,分组(对照组、顺铂组、DMY组、顺铂+DMY组)干预细胞;倒置显微镜观察细胞生长状态;CCK-8检测各组细胞增殖情况;流式细胞术AV/PI双染检测各组细胞凋亡情况;蛋白印迹实验检测凋亡相关蛋白Parp、cleaved Parp、caspase-3、cleaved caspase-3、Bax和Bcl-2表达水平。结果:DMY联合顺铂对细胞的抑制作用显著高于单独应用DMY或顺铂组;流式细胞术AV/PI双染显示联合组的凋亡率显著高于单独用药组;蛋白印记结果显示联合组与单独用药组或与对照组比较总Parp及caspase-3蛋白表达显著降低,cleaved Parp及cleaved Caspase-3蛋白表达显著增加,Bax蛋白水平升高的同时Bcl-2表达减少,Bax/Bcl-2比值显著升高。结论:DMY可以增强顺铂对A549细胞的治疗作用,其机制可能与抑制Parp表达介导的线粒体凋亡有关。OBJECTIVE To explore the effects and mechanism of dihydromyricetin(DMY)on the sensitivity of lung cancer A549 cells to cisplatin so as to provides rationales for chemo-sensitization.METHODS A549 cells cultured in vitro were divided into four groups of control,cisplatin,DMY and cisplatin+DMY.After intervention,cellular growth was observed under an inverted microscope.CCk-8 assay was utilized for detecting cellular proliferation.Flow cytometric AV/PI co-staining assay was employed for detecting apoptosis;The protein expressions of Parp,cleaved Parp,caspase-3,cleaved caspase-3,Bax and Bcl-2 were detected by Western blot.RESULTS The combined group demonstrated the most obvious cellular inhibition.Flow cytometry revealed that apoptosis was much higher in combined group than that in either agent alone group.Western blot indicated that protein expressions of Parp and caspase-3 declined while the expression of protein cleaved Parp and cleaved caspase3 increased.And ratio of Bax/bcl-2 increased in combined group as compared with other groups.CONCLUSION DMY may boost the chemosensitivity of A549 cells to cisplatin.And its mechanism is correlated with a reduction of mitochondrial apoptosis induced by an inhibition of Parp.
