蟹味菇原生质体高压电晕电场诱变的初步研究

摘　　要：以蟹味菇菌丝体为试验材料制备原生质体,并对其进行高压电晕电场诱变处理,以期从中筛选出优质突变菌株。结果显示,经4 kV诱变14 min、6 kV诱变5 min、6 kV诱变8 min、6 kV诱变10 min、8 kV 4 min诱变处理均可获得再生菌落,致死率在69.39%~95.92%,处理条件为4 kV 14min时致死率最低为69.39%。各处理组对比结果显示,高电压短时间处理组更易获得突变菌株,处理电压为6 kV时,处理时间越长菌丝生长速度越快。从再生菌株中筛选出9株单个菌株进行后续试验。结果显示,5号、9号再生菌株生长至2.5 cm所需天数最短为4 d,较对照组缩短了20%的时间,且5号经8 kV诱变4 min的菌株整个培养过程生长速度提高了13.83%。说明对蟹味菇原生质体进行电致诱变处理,可获得生长速度较出发菌株加快的正向突变菌株,为蟹味菇菌种选育提供参考。The mycelium of the Hypsizygus marmoreus was used as experimental material to prepare protoplasts, then treated with high voltage corona field to screen out high quality mutant strains. The results showed that the regenerated colonies could be obtained by mutagenesis of 4 kV 14 min, 6 kV 5 min, 6 kV 8 min, 6 kV 10 min and 8 kV 4 min. The lethality was 69.39%~95.92%, and the lowest lethality was 69.39% when the treatment condition was 4 kV 14 min. Comparison of the two treatment groups revealed that the mutant strains were obtained more easily in the high pressure and short time treatment group. When the treatment voltage keep 6 kV, the longer the treatment time, the faster the mycelium growth. Nine single strains were isolated from the regenerated strains. The time needed for the regenerated strains No.5 and No.9 to grow to 2.5 cm was 4 d, which was20% shorter than that of the control group. Moreover, the growth rate of strain No.5 was increased by 13.83% in the whole culture process after 8 kV 4 min mutation. The results showed that the positive mutant strain could grow faster than the original strain by electro mutagenesis. The results can be used as a reference for the breeding of Hypsizygus marmoreus.
