摘 要:为鉴定羊源腐生葡萄球菌QJ-01菌株并探讨其生物学特性,进行常规生理生化试验、16 S rDNA基因序列分析、小鼠致病性试验、耐药基因检测以及药敏试验。结果显示,分离菌株QJ-01与腐生葡萄球菌生化特性基本一致,16 S rDNA基因扩增获得长度为1465 bp的片段,与GenBank中腐生葡萄球菌的同源性高达99%,在系统进化树中与腐生葡萄球菌(NR041324)聚为一支,结合生理生化特性和16 S rDNA基因序列分析,鉴定该菌株为腐生葡萄球菌。该菌株对小鼠致病性较强,主要损伤肺、脾等脏器。检测出TEM、aac(6′)-Ⅰb、aac(3)-Ⅱa和Sul2共4种耐药基因;该菌株对头孢唑林、环丙沙星和复方新诺明等12种抗菌药物敏感,对青霉素、克林霉素和多黏菌素E等7种抗菌药物耐药。研究结果为进一步探究该菌的致病性及疫病的防治提供参考依据。In order to identify S.S QJ-01 strain isolated from diseased Hu sheep and explore its biological characteristics,in this study,a routine physiological and biochemical tests,16Sr DNA gene sequence analysis,mouse pathogenicity test,drug resistance gene detection and drug sensitivity test were conducted on strain S.S QJ-01.The results showed that the biochemical characteristics of the isolate were basically the same as those of S.staphylococcus.The fragment with length of 1465 bp was obtained by amplification of the 16Sr DNA gene,and the homology with the S.saprophyticus in GenBank was as high as 99%.The phylogenetic tree base on these gene sequences indicated the isolate strain clustered with S.saprophyticus(NR041324).According to physiological and biochemical characteristics as well as the results of 16Sr DNA gene sequence analysis,the isolated strain was identified as S.saprophyticus.The results of pathogenicity test in mice showed that the isolated strain was highly pathogenic and could cause damage to lungs,spleen and other organs.Total four resistance genes were detected from the isolated strain,including TEM,aac(6′)-Ⅰb,aac(3)-Ⅱa and Sul2 genes.The results of drug sensitivity test showed that the isolated strain was sensitive to 11 antimicrobials such as cefazolin,ciprofloxacin and cotrimoxamine,and resistant to 7 antimicrobials such as penicillin,clindamycin and polymyxomycin E.This study provided reference basis for further research on the pathogenicity of S.saprophyticus.
