摘 要:目的探究番茄红素(Lyc)对链脲佐菌素诱导的糖尿病大鼠抗糖尿病作用及机制。方法SPF级雄性SD大鼠60只,试验大鼠适应1 w,其中45只试验大鼠开始连续给予高脂、高糖饮食8 w后,腹腔注射给予链脲佐菌素(STZ)〔60 mg/(kg·d)〕连续2 d制备糖尿病大鼠模型。将造模成功的试验大鼠随机分为3组(n=15):模型组、番茄红素低剂量组(15 mg/kg)和高剂量组(30 mg/kg)。连续给药8 w后测定试验大鼠的体重、空腹血糖,并将试验大鼠处死收集血清和组织样本。测定血清炎症因子和氧化应激水平的变化;采用苏木素-伊红染色法(HE)染色观察肾脏组织的病理学变化。采用Western印迹测定肾脏组织中核因子(NF)-κB p65、p-核因子-κB抑制物激酶(IKK)β、IKκβ、p-IκBα和IKBα蛋白表达量的变化。结果番茄红素能够显著改善糖尿病大鼠的体重、血糖和肾脏指数(P<0.05),并能够显著升高超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性(P<0.05),降低丙二醛(MDA)、肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β和IL-6水平(P<0.05);HE染色结果显示番茄红素对肾脏组织具有显著保护作用,并能够抑制NF-κB p65、p-Iκκβ、IKκβ、p-IκBα和IκBα蛋白表达(P<0.05)。结论番茄红素能够显著抑制糖尿病大鼠血糖升高、炎症和氧化应激损伤,并对肾脏组织具有保护作用,其可能是通过抑制IKK/IKB/NF-κB信号通路来发挥作用的。Objective To investigate the anti-diabetic effect of lycopene on streptozotocin-induced diabetic rats and its mechanism.Methods Sixty male SD rats of SPF grade were given a high-fat and high-sugar diet for 8 weeks, then 60 mg/(kg·d) STZ was injected intraperitoneally for two consecutive days to destroy islet cells to prepare diabetic rat model. The successful experimental rats were randomly divided into three groups(n=15): model group, low-dose lycopene group(15 mg/kg) and high-dose lycopene group(30 mg/kg). Then the body weight and fasting blood sugar of the experimental rats were measured, and the serum and tissue samples were collected. The levels of TNF-α, IL-1β and IL-6 in serum were measured, and the activities of SOD, GSH-Px, CAT and MDA were measured. The HE staining was used to evaluate the pathological changes of kidney in rats. Western blot was used to detect the expression of NF-kappa B p65, p-IKKβ, IKKβ, p-IKBα and IKBα in renal tissue.Results Lycopene could significantly improve the changes of body weight, blood sugar and kidney index in diabetic rats, and could significantly increase SOD, GSH-Px, CAT activity(P<0.05), reduce MDA, TNF-α, IL-1β and IL-6 levels(P<0.05). HE staining showed that lycopene had significant protective effect on kidney tissues, and could significantly increase the activity of SOD, GSH-Px and CAT(P<0.05). It could inhibit the expression of NF-κB p65, p-IKKβ/IKKβ and p-IKBα/IKBα protein(P<0.05).Conclusions Lycopene could significantly inhibit the increase of blood sugar, inflammation and oxidative stress injury in diabetic rats, and has protective effect on kidney tissue. It might play a role by inhibiting IKK/IKB/NF-κB signaling pathway.
