摘  要:目的观察药线灸法对多发梗死性痴呆模型(MID)大鼠海马M受体及环磷酸腺苷反应元件结合蛋白(CREB)含量的影响,分析二者的相关性。方法40只雄性MID大鼠随机分为模型组、药线灸组、药物组和药线非穴组各10只,并取10只正常大鼠为正常组;药线灸组给予“益气调血、扶本培元”药线点灸治疗,药物组给予多奈哌齐灌胃治疗,药线非穴组给予药线点灸两肋下非穴点,模型组与对照组未给予特殊处理,经4 w干预后处死大鼠,分离海马,匀浆、分离上清,检测总M受体、M1、M2受体及CREB含量;用线性相关分析二者关联性,并求回归方程。结果与对照组比较,MID大鼠总M受体、M1受体、CREB含量均显著降低,M2受体显著升高;药线灸组和药物组均能显著升高MID大鼠总M受体、M1受体、CREB的含量,降低M2受体含量;各组总M受体、M1受体与CREB含量呈正相关性,而M2受体与CREB呈负相关性。结论药线灸法可能经由调控ACh-M受体-CREB信息信号通路,从而改善VD认知功能障碍的作用。Objective To observe the effects of "Yiqitiaoxue, fubenpeiyuan" moxibustion of Zhuang medicated thread therapy on mAChR, M1, M2-AChR and CREB content in hippocampus and their correlation in multiple infarct dementia(MID) rats.Methods 40 screened male model rats were randomly divided into model group, the moxibustion of Zhuang medicated thread group, the drug group and the non-acupoint group, 10 rats in each group. Another 10 normal rats were randomly selected into control group. The moxibustion group was treated with the "Yiqi Tiaoxue, Fuben Peiyuan" moxibustion of Zhuang medicated thread therapy, the drug group was treated with donepezil orally, the non-acupoint group was given moxibustion on non-acupoint treatment under the two ribs, and the model group and the control group were given no special treatment. After 4 weeks of intervention, the rats were sacrificed, the hippocampus was isolated, homogenized, and the supernatant was separated. The total mAChR, M1-AChR, M2-AChR and CREB content were detected and counted. Their correlations were analyzed by linear correlation and explored the regression equations. Results Compared with control group, the contents of the total mAChR, M1-AChR and CREB in hippocampal of MID model rats were significantly reduced, while the content M2-AChR was significantly increased(all P<0.05). Both the Zhuang medicated moxibustion group and the drug group showed a significant increasing contents of the total mAChR, M1-AChR, CREB and a significant reducing content of M2-AChR, and the moxibustion group was superior to the drug group in the total mAChR content. There was a positive correlation among the total mAChR, M1-AChR and CREB indicators, but a negative correlation between M2-AChR and CREB indicators in each group. Conclusions "Yiqi Tiaoxue, Fuben Peiyuan" moxibustion of Zhuang medicated thread plays a role in regulating ACh-mAChR-CREB information transmission and signal pathway, and therefore improves vascular dementia cognitive impairment.
