摘  要:目的:分析上调脂蛋白酯酶(LPL)基因对精神分裂症大鼠的干预作用及相关作用机制。方法:选取60只SPF级SD雄性大鼠,分为正常组(正常大鼠)、模型组、下调组、上调组各15只,模型组、下调组、上调组大鼠构建精神分裂症模型,成功后做LPL慢病毒载体滴定,下调组、上调组在大鼠海马组织分别注射含10μL LPL的下调、上调质粒慢病毒悬液。正常组、模型组大鼠注射等量生理盐水。结果:与正常组相比,模型组、下调组LPL基因表达量降低,上调组LPL基因表达量升高(P<0.05)。与下调组相比,上调组刻板行为评分、共计失调评分、平均逃避潜伏期、NO活性、MDA含量、nNOS mRNA及DA水平降低;穿越原平台位置次数,Glu水平,CREB,p-CREB,BDNF,TrkB及p-TrkB蛋白表达量升高(P<0.05)。结论:上调LPL基因具有保护精神分裂症大鼠海马组织的作用,其作用机制可能与激活CREB/BDNF/TrkB信号通路,稳定Glu-DA神经递质系统相关。Objective:To analyze upregulated lipoesterase(LPL)gene intervention and related mechanisms in schizophrenia rats.Methods:Select 60 male SPF grade SD rats,with 15 rats in each of group A(normal rats),group B,group C and group D.After successful LPL lentiviral titration,group C and group D were,respectively,injected with down-regulated and up-regulated plasmid lentiviral suspension containing 10μL LPL.The rats in group A and group B were injected with the same amount of normal saline.After the cpmpletion of the above injection,we observed the outcome of indicators for each group.Results:Compared to group A,group B,group C,the expression of LPL genes in group D was increased(P<0.05).Compared to group C,group D had lower levels of stereotype,total maladjustment,average escape latency,NO activity,MDA content and nNOS mRNA,DA.The number of times of crossing the original platform,the level of Glu and the amount of CREB,p-CREB,BDNF,TrkB,p-TrkB protein wereincreased(P<0.05).Conclusion:Up-regulation of LPL gene can protect the hippocampal tissue for schizophrenia rats,and its mechanism may be related to the activation of CREB/BDNF/TrkB signaling pathway and the stabilization of Glu-DA neurotransmitter system.
