摘  要:通过光谱法和Clark氧电极法研究了重稀土离子铒离子(Er(Ⅲ))对油菜线粒体结构与功能的影响。实验结果表明,Er(Ⅲ)诱导油菜线粒体发生肿胀,其浓度为600μmol·L^(-1)时,油菜线粒体肿胀程度达到最大;Er(Ⅲ)还促进了线粒体内膜对H^(+)/K^(+)的渗透,降低了线粒体膜的流动性。这说明,Er(Ⅲ)可诱导线粒体发生膜渗透性转化(MPT),透射电镜(TEM)观察到500μmol·L^(-1) Er(Ⅲ)作用下的线粒体外膜受损,进一步验证了MPT的发生。Clark氧电极实验结果表明,随着Er(Ⅲ)浓度逐渐增大,线粒体的呼吸好氧速率不断下降,说明Er(Ⅲ)诱导线粒体发生MPT的同时抑制了油菜线粒体的呼吸作用。In this research,the effects of heavy rare earth ions of erbium(Er(Ⅲ))on the structure and function of rape mitochondria were studied by spectroscopy and Clark oxygen electrode method.The experimental results indicated that the Er(Ⅲ)induced the swollen morphology of rape mitochondria.The swollen degree reached the maximum when the ion concentration is up to 600μmol·L^(-1).At the same time,the presence of Er(Ⅲ)enhanced the permeabilization of H^(+)and K^(+)on mitochondrial inner membrane and reduced the fluidity of mitochondrial membranes.These results indicated that Er(Ⅲ)could induce membrane permeability transformation(MPT)in rape mitochondria,and the damage of mitochondrial outer membrane under the concentration of 500μmol·L^(-1) Er(Ⅲ)was observed via transmission electron microscope(TEM),which further verified the occurrence of MPT.Clark oxygen electrode experiment showed that with the concentration of Er(Ⅲ)increasing,the respiratory aerobic rate of mitochondria decreased continuously,which indicated that Er(Ⅲ)can induce MPT in rape mitochondria and inhibited its respiration at the same time.
