摘 要:探索Leptin基因多态性与新疆褐牛肉品质的相关性,旨在寻找与新疆褐牛肉品质性状相关分子标记,为新疆褐牛的选育提供分子遗传学理论依据。利用PCR-RFLP方法检测新疆褐牛Leptin基因第2外显子多态性,并分析其与肉质性状的相关性。结果表明,Leptin基因第2外显子E2JW位点存在AA、AT和TT 3种基因型,处于中度多态,E2FB位点有CT和TT 2种基因型,处于低度多态,均为Hardy-Weinberg非平衡状态。相关分析表明:Leptin基因第2外显子E2JW位点与肌纤维直径呈正相关,且相关关系极显著(r=0.435),与胱氨酸含量正相关,且相关关系显著(r=0.410),与蛋氨酸含量呈弱负相关趋势(r=-0.308)。E2FB位点与硬脂酸含量呈正相关趋势(r=0.425)。结果提示E2JW位点有可能成为影响新疆褐牛肉品质性状的分子遗传标记位点。The association between polymorphisms of Leptin genes and beef quality traits in Xinjiang brown cattle were studied in order to find out molecular markers association with beef quality traits.The polymorphism of Leptin gene’s exon 2 was detected by using PCR-RFLP,and then the correlation of which with beef quality traits was analyzed.The results showed that there were 3 genotypes,i.e.,AA,AT and TT in E2JW of Leptin gene’s exon 2,while 2 genotypes in E2FB,namely CT and TT.E2JW was in moderate polymorphism while E2FB was in low polymorphism.The Leptin polymorphism locus of the population was in a Hardy-Weinberg equilibrium state.Correlation analysis showed that the E2JW locus in Leptin gene’s exon 2 had a intermediate correlation positively with the diameter of muscle fiber(r=0.435),intermediate correlation positively with the content of cystine(r=0.410),and tended to be negatively correlated with the content of methionine(r=-0.308).E2FB locus tended to be positively correlated with stearic acid content(r=0.425).The results indicated that E2JW loucs had a significant association with meat quality traits,and the results of the study will provide a theoretical basis and reference for the molecular marker-assisted selection in Xinjiang brown cattle.
