摘  要:目的:探究青藤碱对胶质瘤细胞SHG-44增殖和侵袭的影响。方法:以0 mmol/L、0.25 mmol/L、0.5 mmol/L、1 mmol/L、2 mmol/L浓度的青藤碱作用于胶质瘤细胞SHG-44,12 h、24 h、36 h后采用CCK-8检测细胞增殖能力,Transwell实验检测胶质瘤SHG-44细胞的侵袭能力,Western blot检测P-JNK、P-ERK和P-STAT3的蛋白表达水平。结果:青藤碱明显抑制胶质瘤SHG-44细胞增殖和侵袭能力,青藤碱抑制胶质瘤细胞增殖的作用与剂量、作用时间相关。青藤碱显著上调胶质瘤SHG-44细胞P-JNK、P-ERK蛋白表达水平,下调P-STAT3蛋白表达水平。结论:青藤碱明显抑制胶质瘤细胞SHG-44增殖和侵袭,可能与调控P-JNK、P-ERK和P-STAT3蛋白表达水平有关。Objective:To investigate the effects of sinomenine on the proliferation and invasion of glioma SHG-44 cells.Methods:The glioma cells were treated with sinomenine at concentrations of 0 mmol/L,0.25 mmol/L,0.5 mmol/L,1 mmol/L and 2 mmol/L and treated with SHG-44 for 12 h,24 h and 36 h.The absorbance was detected by CCK-8 to determine the proliferation ability of the cells.Transwell assay was used to detect the invasion ability of Glioma SHG-44 cells.Western blot was used to detect the protein expression levels of P-JNK,P-ERK and P-STAT3.Results:Sinomenine significantly inhibited the proliferation and invasion of glioma SHG-44 cells.Sinomenine at concentrations of 0.25 mmol/L,0.5 mmol/L,1 mmol/L and 2 mmol/L significantly inhibited the proliferation of glioma cells,and was related to dose and duration of action.Sinomenine significantly upregulated the protein expression of P-JNK,P-ERK and down-regulated P-STAT3 expression in glioma SHG-44 cells.Conclusion:Sinomenine inhibited the proliferation and invasion of Glioma SHG-44 cells,which may be related to the protein expression of P-JNK,P-ERK and P-STAT3.
