摘　　要：目的:探讨PD-1免疫检查点在膀胱癌灌注药物疗效中的作用。方法:常规传代培养人膀胱癌T24细胞,取处于生长对数期的细胞,将其分为对照组、吉西他滨组、吉西他滨+Durvalumab组。采用定量聚合酶链式反应(Q-PCR)检测Caspase-3基因的表达。采用蛋白质印迹法(Western Blot)检测Caspase-3蛋白的表达。采用膜联蛋白V调往检测试剂(AnnexinV-FITC/PI)双染法检测细胞凋亡。采用细胞代谢活性比色法(MTT)检测细胞增殖能力。结果:吉西他滨+Durvalumab组Caspase-3基因的表达水平明显高于吉西他滨组和对照组,且对照组最低;吉西他滨+Durvalumab组Caspase-3蛋白的表达水平明显高于吉西他滨组和对照组,且吉西他滨组与对照组相近;吉西他滨组和吉西他滨+Durvalumab组与对照组相比细胞增殖水平明显降低,且吉西他滨+Durvalumab组为最低;吉西他滨+Durvalumab组凋亡表达水平明显高于吉西他滨组和对照组,而对照组与吉西他滨组比较差异无统计学意义。结论:PD-1主要在各种水平的活化T细胞表面表达,对提升膀胱癌治疗效果具有重要意义。Objective:To explore the role of PD-1 immune checkpoint in the efficacy of drug perfusion for bladder cancer.Methods:Human bladder cancer T24 cells were routinely cultureed,the cells in the logarithmic growth phase were divided into control group,gemcitabine group,gemcitabine+Durvalumab group.Quantitative polymerase chain reaction(Q-PCR)was used to detect the expression of Caspase-3 gene.Western blotting was used to detect the expression of Caspase-3 protein.Adopt Annexin V transfer detection reagent(AnnexinV-FITC/PI)double staining method to detect apoptosis.Cellular metabolic activity colorimetry(MTT)was used to detect cell proliferation ability.Results:The expression level of Caspase-3 gene in gemcitabine+Durvalumab group was significantly higher than that in gemcitabine group and control group,and the lowest in control group.Caspase-3 protein expression level in gemcitabine+Durvalumab group was significantly higher than gemcitabine group and control group,and the gemcitabine group is similar to the control group.The cell proliferation level of the gemcitabine group and the gemcitabine+Durvalumab group was significantly reduced,and the gemcitabine+Durvalumab group was the lowest.The apoptosis expression level of the gemcitabine+Durvalumab group was significantly higher than that of the gemcitabine group and the control group,while there was no significant difference in gemcitabine group and control group.Conclusion:PD-1 is mainly expressed on the surface of activated T cells at various levels,which is of great significance to improve the therapeutic effect of bladder cancer.
