摘  要:目的探讨白细胞介素-1α(interleukin-1α,IL-1α)基因插入/缺失(Ins/Del,I/D)多态性rs3783553与肿瘤易感性的关系。方法截至2020年6月30日,联合应用NCBI PubMed、中国知网(CNKI)、万方和中国生物医学文献数据库(CBM),检索rs3783553与肿瘤易感性相关研究。以合并优势比(OR)及其95%置信区间(CI),估计rs3783553与肿瘤遗传易感性关系。结果共纳入15篇研究,包含肿瘤患者5968例、非肿瘤对照6955例。meta分析结果显示,rs3783553与肿瘤易感性显著关联,在显性模型(Ⅱ/ID vs DD:OR=0.78,95%CI:0.70~0.87)、隐性模型(Ⅱvs DD/ID:OR=0.63,95%CI:0.54~0.73)、纯合子模型(Ⅱvs DD:OR=0.56,95%CI:0.46~0.67)、杂合子模型(ID vs DD:OR=0.85,95%CI:0.77~0.94)和等位基因模型(Ⅰvs D:OR=0.78,95%CI:0.72~0.85)下,Ⅰ等位基因可降低个体罹患肿瘤的风险。结论IL-1αrs3783553 I/D多态性可影响个体对肿瘤的易感性,Ⅰ等位基因是预防肿瘤发生的保护性遗传因素。Objective To investigate the relationship between the insertion/deletion(Ins/del,I/D)polymorphism rs3783553 of interleukin-1α(IL-1α)gene and genetic susceptibility to tumors.Methods The NCBI PubMed,CNKI,Wanfang and Chinese Biomedical Medical Literature Database were searched for case-control studies on the relationship between IL-1αrs3783553 I/D polymorphism and tumor susceptibility up to June 30,2020.Combined odds ratio(OR)and its 95%confidence interval(CI)was used to estimate the relationship between rs3783553 and tumor genetic susceptibility.Results A total of 15 studies with 5968 tumor patients and 6955 non-tumor controls were included.Results of meta-analysis showed that rs3783553 I/D polymorphism was significantly associated with tumor susceptibility,in dominant model(Ⅱ/ID vs DD:OR=0.78,95%CI:0.70-0.87),recessive model(Ⅱvs DD/ID:OR=0.63,95%CI:0.54-0.73),homozygous model(Ⅱvs DD:OR=0.56,95%CI:0.46-0.67),heterozygote model(ID vs DD:OR=0.85,95%CI:0.77-0.94),and allele model(I vs D:OR=0.78,95%CI:0.72-0.85).I allele significantly reduced the risk of tumor in individuals.Conclusion IL-1αrs3783553 I/D polymorphism can affect the susceptibility to tumor,andⅠallele is a protective genetic factor to prevent tumor occurrence.
