摘  要:研究油橄榄叶提取物(Olive leaves extract,OLE)对四种病原菌的抑制作用及抗炎作用.用试管二倍稀释法和牛津杯法测定OLE的抑菌圈直径、最低抑菌浓度(Minimum Inhibitory Concentration,MIC)和最低杀菌浓度(Minimum Bactericidal Concentration,MBC);用小白鼠毛细血管通透试验、小白鼠耳肿胀试验及小白鼠棉球肉芽肿增生试验测定OLE抗炎效果.OLE对大肠埃希氏菌的MIC和MBC为0.125 g/mL和0.25 g/mL,对粪肠球菌和溶血性链球菌的MIC和MBC均为0.0625 g/mL和0.125 g/mL;OLE(1 g/mL)对大肠埃希氏菌、粪肠球菌和溶血性链球菌的抑菌圈直径分别为15.28 mm、17.59 mm和18.37 mm,均为高度敏感,OLE对克雷伯氏肺炎菌无抑制作用.OLE高、中、低剂量组对小白鼠毛细血管通透性增高的抑制率分别是49.15%、35.88%和23.30%;OLE高、中、低剂量组对小白鼠耳肿胀抑制率分别是53.00%、31.34%和9.03%;OLE高、中、低剂量组对小白鼠肉芽肿增生抑制率分别是55.27%、29.39%和23.96%.表明OLE具有一定的抑菌抗炎作用,且抑菌抗炎作用在有限范围内随药物剂量增加而增加.Study on inhibitory and anti-inflammatory effects of Olive Leaves Extract(OLE)on four kinds of bacteria.Tube dilution method and oxford cup method were used to determine inhibition zone,minimum inhibitory concentration(MIC)and minimum bactericidal concentration(MBC),Peritoneal capillary permeability test,Auricle swelling test and cotton-pellet granuloma test in mice were used to observe the anti-inflammatory effect of OLE.The MIC and MBC of OLE against Escherichia coli was 0.125 g/mL and 0.25 g/mL,the MIC and MBC of OLE against Enterococcus faecalis and Hemolytic streptococcus were 0.0625 g/mL and 0.125 g/mL,OLE is highly sensitive against Escherichia coli,Enterococcus faecalis and Hemolytic streptococcusthe,diameters of the inhibition zone were 15.28 mm、17.59 mm and 18.37 mm,OLE had no inhibitory effect on Klebsiella pneumonia.The inhibitory rates of capillaries permeability inhibition were 49.15%、35.88%and 23.30%in high,medium and low dose groups of OLE,the inhibition rates of auricle swelling in high,medium and low dose groups of OLE were 53.00%、31.34%and 9.03%,The inhibitory rates of granuloma hyperplasia were 55.27%、29.39%and 23.96%in high,medium and low dose groups of OLE.The results show that,OLE has highly antibacterial effect against pathogens of Escherichia coli,Enterococcus faecalis and Hemolytic streptococcus,Anti-inflammatory effect and the inhibitory effects are positively correlated with dosages.
