藿苓益智方对阿尔茨海默病模型小鼠神经细胞凋亡线粒体通路的作用研究

摘　　要：目的:探讨藿苓益智方对阿尔茨海默病线粒体通路神经细胞凋亡的调控作用。方法:采用Aβ的活性片段Aβ_(25-35)诱导PC12细胞凋亡建立阿尔茨海默病的细胞模型。本实验设置六个组,即对照组、Aβ_(25-35)组、Aβ_(25-35)+盐酸多奈哌齐组、Aβ_(25-35)+藿苓益智方提取物(50、100、200μg/ml)组。利用流式细胞仪、荧光酶标仪和透射电镜等技术,检测霍苓益智方提取物对阿尔茨海默病细胞模型中细胞存活率、神经细胞凋亡率、线粒体超微结构的变化。结果:藿苓益智方提取物50μg/ml组的细胞存活率与Aβ_(25-35)组相比有一定的增高(P<0.05);盐酸多奈哌齐组和藿苓益智方提取物(100、200μg/ml)组与Aβ_(25-35)组相比,细胞存活率显著升高(均P<0.01),但藿苓益智方提取物200μg/ml组升高的更明显;藿苓益智方低、中、高剂量+A _(25-35)组细胞凋亡率则逐步下降,其中中、高剂量组与Aβ_(25-35)组相较差异均具有统计学意义(均P<0.01);藿苓益智方低、中、高剂量+Aβ_(25-35)组对细胞核的损害程度上,经治疗以后逐步缓解,且呈剂量依赖性趋势。结论:藿苓益智方对阿尔茨海默病的治疗中可对Aβ诱导的PC12细胞凋亡具有明显的抑制作用,改善细胞存活率,减少线粒体细胞凋亡。Objective:To investigate the regulatory effect of Huoling Yizhi recipe on apoptosis of neurons in mitochondrial pathway.Methods:The apoptosis of PC12 cells was induced by Aβ_(25-35),the active fragment of Aβ,to establish the cell model of Alzheimer disease.In this experiment,six groups were set up:control group,Aβ_(25-35) group,Aβ_(25-35)+donepezil hydrochloride group,Aβ_(25-35)+Huoling Yizhi extract(50,100,200μg/ml)group.The changes of cell survival rate,neuron apoptosis rate and mitochondrial ultrastructure in Alzheimer disease cell model were detected by flow cytometry,fluorescent enzyme labeling and transmission electron microscopy.Results:Compared with Aβ_(25-35) group,the cell survival rate of the 50μg/ml group was higher(P<0.05);Compared with the Aβ_(25-35) group,the cell survival rate of the donepezil group and the 100,200μg/ml group was significantly higher(all P<0.01),but the 200μg/ml group was more significantly higher;the low,medium and high dose+Aβ_(25-35) group the apoptotic rate decreased gradually,and there was a significant difference between the middle and high dose groups and Aβ_(25-35) group(all P<0.01);The degree of nuclear damage in the low,middle and high dose+Aβ_(25-35) group of Huoling Yizhi ricipe was gradually relieved after treatment,and showed a dose-dependent trend.Conclusion:In the treatment of Alzheimer disease,Huoling Yizhi recipe can obviously inhibit the apoptosis of PC12 cells induced by Aβ,improve the cell survival rate and reduce the apoptosis of mitochondrial cells.
