华蟾素片对CT26结肠腺癌诱导恶病质小鼠肌肉萎缩的影响

摘　　要：目的探讨华蟾素片对CT26结肠腺癌诱导的恶病质小鼠净体质量、肌肉萎缩等方面的影响。方法建立CT26结肠腺癌细胞恶病质小鼠模型,24只小鼠随机分为正常组、单纯恶病质组、实验组。测定对比小鼠实验后净体质量、心肌、腓肠肌、附睾脂肪质量;RT-PCR法测定对比实验后腓肠肌MAFbx和Mu RF-1 mRNA表达。结果实验前后小鼠的净体质量的对比,单纯恶病质组下降最为明显,差异有统计学意义(t=4.055,P<0.05)。实验后单纯恶病质组小鼠腓肠肌MAFbx和Mu RF-1 mRNA表达为(97.68±6.21)、(36.77±3.74)mg,而实验组为(57.24±6.29)、(26.79±3.73)mg,两组相比实验组的骨骼肌萎缩明显缓解,差异有统计学意义(P<0.05)。结论对于癌性恶病质小鼠模型,经华蟾素片的干预,能够明显减缓其肌肉的萎缩速度。Objective To investigate the effects of Huachansu tablets on the net body weight and muscle atrophy of cachexia mice induced by CT26 colon adenocarcinoma.Methods A mouse model of CT26 colon adenocarcinoma cachexia was established.24 mice were randomly divided into normal group,simple cachexia group,and experimental group.The net body mass,myocardium,gastrocnemius,and epididymal fat mass of the mice after the experiment were measured and compared;the expression of MAFbx and Mu RF-1 mRNA in the gastrocnemius muscle after the comparison experiment was measured by RT-PCR.Results Compared with the net body weight of the mice before and after the experiment,the pure cachexia group had the most significant decrease,and the difference was statistically significant(t=4.055,P<0.05).After the experiment,the expression of MAFbx and Mu RF-1 mRNA in gastrocnemius of the simple cachexia group was(97.68±6.21)mg,(36.77±3.74)mg,while that of the experimental group was(57.24±6.29)mg,(26.79±3.73)mg.Compared with the experimental group,the two groups had atrophy of skeletal muscle significantly relieved,the difference was statistically significant(P<0.05).Conclusion For the mouse model of cancer cachexia,the intervention of Huachansu tablets can significantly slow down the rate of muscle atrophy.
