摘  要:目的:探究氟西汀预处理对脑缺血再灌注(I/R)小鼠DNA损伤修复蛋白Ku80和XRCC1的影响。方法:将120只小鼠,随机分为假手术组(Sham)、缺血再灌注组(I/R)和氟西汀组(Fluo),每组各40只。采用线栓法建立I/R小鼠模型,在术前1周给予连续灌胃氟西汀[10 mg/(kg·d)]处理,其他组给予相同量的生理盐水灌胃。在再灌注2 h、12 h、24 h和48 h分别进行TTC染色,通过Western blot测定DNA损伤修复蛋白定位和变化。结果:与Sham组相比,再灌注24 h脑梗死面积出现明显增加,再灌注2 h DNA损伤及修复蛋白Ku80和XRCC1表达明显下调。与I/R组相比,Fluo组在灌注12 h、24 h和48 h脑梗死面积明显减小,再灌注2 h开始DNA损伤及修复蛋白Ku80和XRCC1表达明显上调。结论:氟西汀预处理后,再灌注小鼠海马DNA损伤修复蛋白Ku80和XRCC1表达增加,改善了再灌注小鼠海马DNA损伤修复功能。Objective To investigate the effects of fluoxetine preconditioning on DNA damage repair proteins KU80 and XRCC1 in mice with cerebral ischemia reperfusion(I/R).Method 120 mice were randomly divided into Sham group(Sham),I/R group(I/R)and fluoxetine group(Fluo).40 mice in each group.The mouse Model was established by suture method.Fluoxetine[10 mg/(kg·d)]was given continuously one week before operation,and the other groups were given the same amount of normal saline.TTC staining was performed at 2 h,12 h,24 h and 48 h respectively the localization and changes of DNA damage repair proteins were determined by western blot.Results Compared with Sham group,the infarct size was significantly increased after 24 h reperfusion.The expression of KU80 and XRCC1 were significantly decreased after 2 h reperfusion.Compared with I/R group,the area of cerebral infarction was significantly decreased at 12 h,24 h and 48 h.The expression of KU80 and XRCC1 was up-regulated at 2 h after reperfusion.Conclusion The expression of KU80 and XRCC1 in hippocampus DNA damage repair protein was increased after fluoxetine pretreatment,which improved the function of hippocampus DNA damage repair.
