摘  要:目的探讨大鼠骨髓间充质干细胞(BMSC)对缺血性脑卒中小胶质细胞类型是否有逆转作用。方法选择成功构建大脑中动脉阻塞模型SD大鼠20只,随机选2只做TTC染色测脑梗死灶,另18只分为模型组和BMSC组(侧脑室定位注射5μl含BMSC 10~5个),每组9只,2组分别于1、3和7 d各取3只大鼠断头取材,脑组织切片用离子钙结合衔接分子(Iba)和抗CD206或TNF-α双重免疫荧光染色,计数双染重合阳性细胞。结果TTC染色示,模型大鼠脑梗死病灶呈苍白色。双重免疫荧光染色示,与模型组比较,BMSC组1、3和7 d脑缺血区Iba+CD206双染细胞阳性数明显增加(P=0.000),3和7 d脑缺血区Iba+TNF-α双染细胞阳性数明显减少[(42.22±9.64)个/mm^(2)vs(86.43±12.68)个/mm^(2),(42.67±13.06)个/mm^(2)vs(88.89±13.70)个/mm^(2),P=0.000]。结论大脑中动脉阻塞模型大鼠注入BMSC后,脑缺血区的抗炎型小胶质细胞增加,促炎型小胶质细胞减少。Objective To study whether BMSC can reverse the types of microgliacytes in ischemic stroke rats.Methods Twenty SD rats were included in this study.After a middle cerebral artery occlusion model was established,the infarction sites in 2 rats randomly selected were detected with TTC staining.The remaining 18 SD rats were divided into model group(n=9)and BMSC group(n=9).Brain tissue samples were taken from 3 rats of model group and BMSC group respectively on days 1,3,7 and stained with anti-Iba and anti-CD206 or TNFαdouble immunofluorescence.The doubly-stained overlap positive cells were calculated and compared.Results TTC staining showed the white cerebral infarction in model group.Double immunofluorescence staining displayed that the number of Iba+CD206 double positive cells was significantly larger in BMSC group than in model group on days 1,3 and 7(P=0.000)while that of Iba+TNFαdouble positive cells was significantly smaller on days 3 and 7(42.22±9.64/mm^(2)vs 86.43±12.68/mm^(2),42.67±13.06/mm^(2)vs 88.89±13.70/mm^(2),P=0.000).Conclusion The number of antiinflammatory microgliacytes is increased while that of proinflammatory microgliacytes is deceased after BMSC are injected into the middle cerebral artery occlusion model of rats.
