摘　　要：目的:探讨微小染色体维持蛋白6(MCM6)在肾透明细胞癌(ccRCC)早期诊断及预后判断中的意义。方法:应用TCGA数据库中533例ccRCC患者的RNA测序(RNAseq)数据,从RNA水平初步分析MCM6的mRNA表达与ccRCC早期诊断及预后的关系。应用免疫组化(IHC)实验方法从蛋白质层面分析MCM6蛋白在373例ccRCC患者的表达与ccRCC早期诊断与预后的关系。结果:分析TCGA数据库发现,ccRCC患者的MCM6高表达与肿瘤进展和生存率低均有关(P<0.001)。对373例ccRCC组织的免疫组化分析表明MCM6蛋白在ccRCC的癌旁正常组织中几乎不表达,而在ccRCC的肿瘤细胞中明显表达。Kaplan-Meier生存分析显示MCM6蛋白是ccRCC患者生存的不利因素,高表达MCM6的ccRCC患者预后差(P<0.001)。Cox回归分析结果显示,MCM6是ccRCC的独立预测因子(P<0.001)。MCM6受试者工作特征曲线下面积(AUC)值为0.604(P=0.001)。结论:MCM6是ccRCC患者生存的不利因素,MCM6是一种潜在的ccRCC早期诊断及判断预后的新型标志物。Objective:To investigate the significance of microchromosome maintenance protein 6(MCM6)in early diagnosis and prognosis of clear cell renal cell carcinoma(ccRCC).Method:Using the RNA sequencing(RNAseq)data of 533 ccRCC patients in the TCGA database,the relationship between the mRNA expression of MCM6 and the early diagnosis and prognosis of ccRCC from the RNA level was analyzed.Then immunohistochemical(IHC)method was used to analyze the relationship between the expression of MCM6 protein and the early diagnosis and prognosis of ccRCC in 373 patients with ccRCC.Result:Analysis of the TCGA database showed that the high expression of MCM6 in patients with ccRCC was associated with low tumor progression and low survival rate(P<0.001).The immunohistochemical analysis of 373 cases of ccRCC tissues proved that MCM6 protein was almost not expressed in the adjacent normal tissues of ccRCC,but was expressed in tumor cells of ccRCC.Kaplan-Meier survival analysis showed that MCM6 protein was an unfavorable factor for the survival of ccRCC patients,and the prognosis of ccRCC patients with high expression of MCM6 was poor(P<0.001).Cox regression analysis showed that MCM6 was an independent predictor of ccRCC(P<0.001).The area under receiver operating characteristic curve(AUC)of MCM6 was 0.604(P=0.001).Conclusion:MCM6 is an unfavorable factor for the survival of patients with ccRCC,MCM6 is a potential marker for early diagnosis and prognosis of ccRCC.
