摘 要:目的:探讨癌-睾丸抗原OY-TES-1和丙酮酸激酶M2型(PKM2)在人肝细胞癌(HCC)中的表达,并分析OY-TES-1对PKM2表达的影响。方法:利用LinkedOmics数据库筛选OY-TES-1表达正相关基因集;使用DAVID数据库对OY-TES-1进行功能富集分析,了解HCC中OY-TES-1潜在的生物学功能,寻找功能相关基因;通过GEPIA数据库分析和实时荧光定量PCR(qPCR)实验检测,探讨OY-TES-1和功能相关基因在HCC中的表达及临床意义;干扰HCC细胞株中OY-TES-1的表达,qPCR和Western blotting检测OY-TES-1功能相关基因的表达变化。结果:LinkedOmics数据库筛选获得1001个与OY-TES-1表达正相关的基因集;功能富集结果显示,OY-TES-1可能通过PKM2参与HCC细胞多糖合成、脂代谢、碳水化合物代谢等信号通路。GEPIA数据库分析显示,HCC组织中OY-TES-1和PKM2的表达显著高于正常肝组织,两者表达呈正相关关系(r=0.3;P<0.001);qRT-PCR证实OY-TES-1(癌组织:0.58±0.95 vs.癌旁组织:0.36±0.61,P<0.05)和PKM2(癌组织:0.14±0.17 vs.癌旁组织:0.08±0.14,P<0.05)在HCC组织中均为高表达,两者的表达呈正相关关系(r=0.482,P<0.001);尚未发现OY-TES-1和PKM2的表达与HCC患者临床参数相关。在HepG2和BEL-7404细胞株中干扰OY-TES-1表达,PKM2的mRNA和蛋白表达随之明显下调。结论:OY-TES-1和PKM2均高表达于HCC,且两者具有较强的表达相关性;HCC中OY-TES-1能够影响PKM2的表达。Objective:To explore the expression of cancer-testis antigen OY-TES-1 and pyruvate kinase isozyme type M2(PKM2)in human hepatocellular carcinoma(HCC),and to analyze the effect of OY-TES-1 on the expression of PKM2.Methods:The LinkedOmics database was used to screen the positive correlation gene set of OY-TES-1.Functional enrichment analysis of OY-TES-1 was performed using the DAVID database to understand the potential biological functions of OY-TES-1 and obtain function-related genes in HCC.Through GEPIA database analysis and qRT-PCR assay,the expression and clinical significance of OY-TES-1 and its function-related gene in HCC were investigated.Interfering with the expression of OY-TES-1 in HCC cell lines,qRT-PCR and Western blottingwere used to detect the expression changes of OY-TES-1 function-related genes.Results:The set of 1,001 genes positively correlated with OY-TES-1 was obtained by the LinkedOmics database.OY-TES-1 may participate in polysaccharide synthesis,lipid metabolism,carbohydrate metabolism,and other signaling pathways through PKM2.GEPIA database analysis showed that the expression of OY-TES-1 and PKM2 in HCC was significantly higher than that in normal liver tissues,and both of them had a positive correlation(r=0.3;P<0.001).qRT-PCR confirmed that OY-TES-1(tumor:0.58±0.95 vs.adjacent:0.36±0.61,P<0.05)and PKM2(tumor:0.14±0.17 vs.adjacent:0.08±0.14,P<0.05)were highly expressed in HCC,and both of them had a strong expression correlation(r=0.482,P<0.001).Statistical analysis showed that the expression of OY-TES-1 and PKM2 did not correlate with the clinical parameters of HCC patients.After interfering with the expression of OY-TES-1 in HepG2 and BEL-7404 cell lines,the mRNA and protein expression of PKM2 weresignificantly down-regulated.Conclusion:OY-TES-1 and PKM2 are highly expressed in HCC and have a strong expression correlation.OY-TES-1 can affect the expression of PKM2 in HCC.
