摘  要:为鉴定猪乳中的蛋白质组分,研究采集了8头大白猪的全乳,经离心脱脂后,采用SDS-PAGE对脱脂乳中的蛋白质进行分离,然后切取凝胶上的3条蛋白质条带,经还原烷基化和胰蛋白酶解处理后,采用液相色谱-串联质谱分析产生的水解肽,以鉴定这3种低分子量蛋白质。结果表明,获得鉴定的2种蛋白质包括β-乳球蛋白(β-LG)和乳清酸蛋白(WAP),其中WAP可能对猪乳的生物学功能有重要价值。In order to identify several protein components of porcine milk,whole milk samples of large white pigs(n=8)were collected,and skim milk was obtained by centrifugation.SDS-PAGE was employed to separate skim milk proteins,and tree protein bands on SDS-gel were cut off,followed by treatments of reductive alkylation and trypsin digestion.The resulting peptides were loaded on LCMS/MS to identify the three proteins with low molecular weight.Two proteins includingβ-lactoglobulin(β-LG)and whey acidic protein(WAP)were identified successfully.WAP may probably affect porcine milk functions.
