摘  要:目的观察舒芬太尼(Sufentanil,SUF)对神经性疼痛大鼠的神经阻滞作用,探讨Nrf-2/HO-1信号通路的调控机制。方法 SD大鼠随机分为假手术组(Sham)、坐骨神经结扎法建立神经性疼痛模型组(chronic constriction injury,CCI)、CCI+舒芬太尼组(SUF),每组10只。测定各组大鼠机械性缩足反射阈值和热缩足潜伏期;试剂盒检测大鼠坐骨神经中超过氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)含量;Western blot检测大鼠坐骨神经中Bax、Bcl-2、Cleaved-caspase-3、Nrf-2及HO-1蛋白的表达。结果与CCI组比较,SUF显著提高CCI大鼠的机械痛阈值和热缩足潜伏期,显著增加坐骨神经中SOD、GSH-Px活性并降低MDA含量,降低Bax/Bcl-2比值及Cleaved-caspase-3蛋白的表达水平。结论 SUF改善CCI诱导的神经病理性疼痛,抑制坐骨神经氧化应激及细胞凋亡,与激活Nrf-2/HO-1信号通路相关。Objective To observe the effect of Sufentanil(SUF) on the nerve block of neuropathic pain rats, and to explore the regulatory mechanism of Nrf-2/HO-1 signaling pathway. Methods SD rats were randomly divided into sham group, chronic constriction injury(CCI,neuropathic pain model group by ligation of sciatic nerve), and CCI+sufentanil group(SUF). The mechanical withdrawal threshold and thermal withdrawal latency was measured, the activities of excess oxidizing dismutase(SOD) and glutathione peroxidase(GSH-Px), and the content of malondialdehyde(MDA) in the sciatic nerve were determined by biochemistry methods. The expressions of Bax, Bcl-2, Cleaved-caspase-3, Nrf-2 and HO-1 proteins in the sciatic nerve were detected by Western blot. Results Compared with the CCI group, SUF significantly increased the mechanical pain threshold and heat shrinkage latency of CCI rats, significantly increased the activity of SOD and GSH-Px and reduced the content of MDA, downregulated the ratio of Bax/Bcl-2 and Cleaved-caspase-3 protein expression level in the sciatic nerve. Conclusion SUF improved CCI-induced neuropathic pain, inhibit sciatic nerve oxidative stress and apoptosis via activating Nrf-2/HO-1 signaling pathway.
