摘  要:目的探讨WWC3在胶质瘤干细胞(GSCs)中的表达,以及通过调节Hippo通路调控GSCs增殖、迁移和侵袭的作用。方法 Western blot检测Hippo通路相关分子的表达。过表达WWC3后,检测MST1、LATS1和YAP的磷酸化与表达;免疫荧光检测YAP分布变化;CCK8和Transwell检测GSCs增殖、迁移和侵袭的变化。结果 WWC3、MST1和LATS1在GSCs中的表达均显著降低,而YAP的表达显著升高。过表达WWC3显著增加p-MST1/MST1、p-LATS1/LATS1、p-YAP/YAP的表达水平,抑制细胞核中YAP表达,抑制GSCs的增殖、迁移和侵袭。结论过表达WWC3通过降低Hippo通路活性抑制GSCs的增殖、迁移和侵袭。Objective To investigate the expression of WWC3 in glioma stem cells(GSCs) and the role of regulating the proliferation, migration, and invasion of GSCs by regulating the Hippo pathway. Methods Western blot was conducted to detect the expression of related molecules in Hippo pathway. The phosphorylation and expression of MST1, LATS1, and YAP were respectivelydetected after overexpressing WWC3. The change of YAP distribution was detected by Immunofluorescence. CCK8 and Transwell assaywere respectively conducted to detect the changes of GSCs proliferation, migration, and invasion. Results The expression of WWC3, MST1, and LATS1 in GSCs were all significantly reduced, while the expression of YAP was significantly increased. Overexpression of WWC3 significantly increased p-MST1/MST1, p-LATS1/LATS1, and p-YAP/YAP, suppressed the expression of YAP in the nucleus, and inhibited the proliferation, migration, and invasion of GSCs. Conclusion Overexpression of WWC3 inhibited the proliferation, migration, and invasion of GSCs by reducing the activity of the Hippo pathway.
