摘 要:试验旨在探究肠炎沙门氏菌攻毒对SPF雏鸡粪便微生物区系的影响。选择1日龄SPF雏鸡24只,随机分为2组,每组12只。在4日龄处理组口服107 CFU肠炎沙门氏菌(ATCC13076)攻毒,对照组口服等量PBS溶液。试验期22 d。结果显示:①处理组SPF鸡的存活率为75%,对照组全部存活,期间两组鸡的体重无显著性差异;②OTU水平上,相同日龄两组鸡粪便菌群的丰富度相近,中期处理组粪便菌群多样性更高(P<0.05),后期对照组菌群稳定性更高,而且攻毒对SPF鸡粪便菌群的影响随日龄增加而减弱;③在属水平上,与对照组相比,处理组Bacteroides(拟杆菌门)、Enterococcus(肠球菌属)和Aeriscardovia(卡多维亚氏菌属)的相对丰度明显升高(P<0.05);Lactobacillus(乳杆菌属)、Alistipes(另枝菌属)、Faecalibacterium(粪杆菌属)、Romboutsia(罗姆布茨菌属)和Ruminococcaceae_UCG_005(瘤胃球菌UCG_005)的相对丰度明显降低(P<0.05);④KEGG分析显示攻毒影响了菌群参与的多个代谢通路。综上,肠炎沙门氏菌感染影响了SPF雏鸡粪便菌群的结构组成、稳定性及菌群参与的代谢通路。Bacteroides(拟杆菌门)、Lactobacillus(乳杆菌属)等差异菌属可能对SPF雏鸡抵抗沙门氏菌入侵起到积极作用。The purpose of the experiment was to explore the impact of Salmonella enteritidis challenge on the fecal microflora of SPF chicks.Totally 241-day-old SPF chicks were randomly divided into two groups with 12 each group.The treatment group was orally challenged with 0.2 ml(approximately 107 CFU)of Salmonella enteritidis(ATCC13076)at the age of 4 days,while the control group was given equal volume of sterile PBS solution.The trial lasted for 22 days.The results showed that:①All SPF chicks survived in the control group,and the survival rate was 75%in the treatment group.There was no significant difference in body weight between the two groups during the experiment;②At the OTU level,the abundance of fecal flora of the two groups was similar at the same age,the diversity of the flora in the treatment group in the middle of the experiment was higher(P<0.05),and the stability of the flora in the control group was higher in the later period of the experiment.Moreover,the impact of the challenge on the fecal flora of SPF chickens became weaker with increasing age;③Compared with the control group,the relative abundance of Bacteroides,Enterococcus and Aeriscardovia in the treatment group was significantly increased(P<0.05);however,the relative abundance of Lactobacillus,Alistipes,Faecalibacterium,Romboutsia and Ruminococcaceae_UCG_005 was significantly decreased(P<0.05);④KEGG analysis showed that the challenge greatly affected the metabolic pathways that the fecal flora participates in.In conclusion,Salmonella enteritidis infection can affect the structure and stability of fecal flora in SPF chicks,and change the metabolic pathways that the flora participates in.Differential bacteria including Bacteroides and Lactobacillus,may play a positive role in the resistance of SPF chicks to the invasion of Salmonella.
