摘 要:目的探究帕金森病(PD)模型中p150^(Glued)蛋白的表达及分布。方法选取20只雄性C57/BL6成年小鼠随机分为PD小鼠模型组[腹腔注射1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP),20mg/kg]和空白对照组(腹腔注射与模型组等体积生理盐水),每组10只。免疫荧光染色检测两组小鼠中脑黑质致密部酪氨酸羟化酶(TH)和p150^(Glued)的表达差异。另选取MN9D细胞分为PD细胞模型组[不同浓度的1-甲基-4-苯基吡啶离子(MPP+)作用细胞48h]和空白对照组(加等体积的磷酸缓冲盐溶液替代),利用MTT比色法检测MN9D细胞存活率,蛋白质印迹技术(Western blot)及免疫荧光染色检测p150^(Glued)在细胞中的表达。结果在PD小鼠模型中,与对照组相比,黑质区域TH阳性神经元细胞数量减少,p150^(Glued)表达量降低。在PD细胞模型中,p150^(Glued)在胞体中异常聚集,其聚集体与微管蛋白共定位。与对照组相比较,MPP+可下调MN9D细胞中p150^(Glued)蛋白的表达,并随着MPP+的浓度增加,p150^(Glued)蛋白表达降低,差异有统计学意义(P<0.05)。结论p150^(Glued)可能是诱导多巴胺神经元凋亡及影响PD发病的一个重要因素。Objective To investigate the expression and distribution of p150^(Glued) protein in Parkinson’s disease(PD)model.Methods Twenty adult male C57/BL6 mice were selected and randomly divided into PD model group[intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP),20 mg/kg]and blank control group(intraperitoneal injection of the same volume of normal saline as the model group),with 10 mice in each group.The expression differences of tyrosine hydroxylase(TH)and p150^(Glued) in the dense part of the substantia nigra in the midbrain of the two groups were detected by immunofluorescence staining.The MN9 Dcells were divided into PD cell model group[treated with different concentrations of 1-methyl-4-phenylpyridine(MPP+)for48 h]and blank control group(equal volume of phosphate buffer).The survival rate of MN9 Dcells was detected by MTT colorimetry.The expression of p150^(Glued) in cells was detected by Western blot and immunofluorescence staining.Results In PD mouse model,the number of TH positive neurons in the substantia nigra region decreased,and the expression of p150^(Glued) decreased,compared with the blank control group.In PD cell model,p150^(Glued) aggregated abnormally in the cell body,and its aggregates were co-localized with tubulin.Compared with the control group,MPP+could down-regulate the expression of p150^(Glued) protein in MN9 Dcells,and the expression of p150^(Glued) protein decreased with the increase of MPP+concentration,and the difference was statistically significant(P<0.05).Conclusion p150^(Glued) may be the induction of dopamine neurons apoptosis and an important factor affecting the pathogenesis of PD.
