摘 要:目的观察滇白珠乙酸乙酯提取物(简称滇白珠提取物)对大肠及小肠性腹泻小鼠的止泻作用,并观察其可能的胃肠功能调节机制。方法采用番泻叶和蓖麻油诱导建立大肠及小肠性腹泻小鼠模型,分别按照体质量灌胃给予20、10、5 g/kg(按生药计算)的滇白珠提取物及6 mg/kg易蒙停、等体积溶媒(3.2%吐温-80溶液)进行预处理,记录小鼠的稀便率、稀便级及腹泻指数。取正常昆明小鼠及胃肠功能亢进小鼠(皮下注射甲硫酸新斯的明0.15 mg/kg),采用炭末推进实验观察20、10、5 g/kg滇白珠提取物及6 mg/kg易蒙停、等体积溶媒灌胃处理对其胃肠推进运动的影响,比较其胃内残留率、小肠推进率。取SPF级SD大鼠离体小肠进行张力试验,记录终浓度分别为44.78、22.39、11.19μg/mL的滇白珠提取物及硫酸阿托品、溶媒作用2.5、5.0 min的大鼠离体正常小肠张力抑制率,终浓度分别为89.11、44.56、22.28μg/mL的滇白珠提取物及硫酸阿托品、溶媒作用2.5、5.0 min的大鼠离体张力亢进小肠(加入15 mg/L氯化乙酰胆碱)张力抑制率。结果在大肠及小肠性腹泻小鼠中,20、10 g/kg滇白珠提取物及易蒙停作用后小鼠稀便率、稀便级及腹泻指数均较溶媒作用后降低,5 g/kg滇白珠提取物作用后小鼠腹泻指数均较溶媒作用后降低(P均<0.01)。与溶媒作用后比较,20、10 g/kg滇白珠提取物及易蒙停作用后正常小鼠小肠推进率均降低,胃肠功能亢进小鼠胃内残留率均升高、小肠推进率均降低(P均<0.05)。与溶媒作用后相同时间点比较,加入不同终浓度滇白珠提取物及硫酸阿托品作用2.5、5.0 min的大鼠离体正常小肠及张力亢进小肠张力均降低、张力抑制率均升高(P均<0.05)。结论滇白珠提取物对大肠及小肠性腹泻小鼠均具有明显的止泻作用,其机制可能与通过拮抗M胆碱能受体而增加胃内残留率、降低小肠推进速度、解除胃肠道平滑肌痉�Objective To study the anti-diarrhea effect of the ethyl acetate extract of gaultheria leucocarpa(APGL)and to observe its regulatory mechanism of gastrointestinal function.Methods Two models of diarrhea were established,which were induced by folium sennae and castor oil.Then,20,10,and 5 g/kg of APGL(calculated by crude drug),6 mg/kg of imodium or equal volume solvent(3.2 % Twain-80 solution) were given intragastrically to the related mice according to the body weight,respectively.The loose stool rate,loose stool grade and diarrhea index of mice were recorded.Terminal carbon propulsion test was carried out to observe the effects of 20,10 and 5 g/kg of APGL,6 mg/kg of imodium or equal volume solvent on gastrointestinal propulsion of normal mice and hyper gastrointestinal function mice(injected with 0.15 mg/kg of neostigmine methylsulfate).The gastric residual rate and intestinal propulsion rate were compared.In the tension test of small intestine of SPF grade SD rats,the inhibitory rates of the isolated normal small intestine were recorded with treatments of 44.78,22.39 and 11.19 g/mL of APGL,atropine sulfate or solvent for 2.5 and 5.0 min,the inhibitory rates of the isolated hyper small intestine(added with 15 mg/L acetylcholine chloride) were recorded with treatments of 89.11,44.56 and 22.28 g/mL of APGL,atropine sulfate or solvent for 2.5 and 5.0 min,respectively.Results In two models of diarrhea mice,the loose stool rate,loose stool grade and diarrhea index were decreased after the treatments of APGL(20,10 g/kg) and imodium(6 mg/kg),compared with the solvent treatment(all P<0.01).The diarrhea indices in two models were decreased after the treatment of APGL(5 g/kg),compared with the solvent treatment(all P<0.01).Compared with the solvent treatment,the small intestinal propulsive rate of normal mice was decreased,while the gastric residual rate was increased and the small intestinal propulsive rate was decreased in hypergastrointestinal mice,with the treatments of APGL(20,10 g/kg) and imodium(6 mg/kg)(all P<0.05)
