摘 要:目的通过生物信息学方法,探索影响上皮性卵巢癌腹腔转移的关键基因及信号通路。方法自基因表达数据库(GEO)下载数据集GSE30587,通过在线工具GEO2R分析获取差异基因,通过DAVID数据库进行GO和KEGG分析,通过STRING数据库分析制作基因之间PPI网络。结果上皮性卵巢癌原发灶与网膜转移灶存在60个表达差异的基因。GO和KEGG分析发现,差异基因常与细胞外基质成分等功能或通路相关,在PPI网络上,COL1A1、COL3A1、COL5A2、COL6A3、MMP2、FN1、DCN基因可能在上皮性卵巢癌腹腔内转移中起到关键作用。结论通过这些芯片数据的生物信息学分析,筛选了与上皮性卵巢癌腹腔内转移密切相关的基因和通路,其中COL1A1、COL3A1、COL5A2、COL6A3、MMP2、FN1、DCN等细胞外基质基因及通路可能在上皮性卵巢癌腹腔内转移中发挥重要作用,期待进一步深入研究。Objective To explore the key genes and signaling pathways that affect the metastasis of epithelial ovarian cancer in the abdominal cavity through bioinformatics methods.Methods Download the data set GSE30587 from the public database GEO,differential genes were obtained by using the online tool GEO2R analysis,conducted GO and KEGG analysis through the DAVID database,and create the PPI network between genes through the STRING database analysis.Results There were 60 differently expressed genes between primary lesions of ovarian cancer and omental metastases lesions.GO and KEGG analysis found that differential genes are often related to functions or pathways such as extra-cellular matrix components.On the PPI network,COL1A1,COL3A1,COL5A2,COL6A3,MMP2,FN1,and DCN genes may be involved in intra-abdominal metastasis of epithelial ovarian cancer.Conclusions Through the bioinformatics analysis of these chip data,the genes and pathways closely related to intra-abdominal metastasis of epithelial ovarian cancer were screened,including COL1A1,COL3A1,COL5A2,COL6A3,MMP2,FN1,DCN and other extra-cellular matrix genes and pathways.They may play an important role in intra-abdominal metastasis of epithelial ovarian cancer,and we look forward to further research.
