摘 要:目的:研究长链非编码RNA(Long non-coding RNA,LncRNA)MEG3对乳腺癌细胞侵袭及迁移的影响并探究其机制。方法:通过实时荧光定量PCR(qRT-PCR)检测乳腺癌细胞MDA-MB-231、MCF-7、SKBR-3中LncRNA MEG3表达,选取表达水平较低的MCF-7细胞株,构建pcDNA3.1-MEG3重组质粒,过表达MCF-7中的LncRNA MEG3,再通过qRT-PCR检查细胞miRNA-21变化情况。其次,通过划痕实验检测MCF-7细胞迁移变化;Transwell实验检测MCF-7细胞侵袭变化;Western blot检测上皮间质转化(EMT)相关分子(E-Cadherin、N-Cadherin、Vimentin)的表达量变化。结果:在MEG3过表达的MCF-7细胞株中,miRNA-21表达量降低,且细胞的侵袭及迁移能力也下降,细胞中的E-Cadherin蛋白表达量增高,而N-Cadherin及Vimentin表达量降低,差异均有统计学意义(P均<0.05)。结论:LncRNA MEG3通过调控miRNA-21影响乳腺癌细胞侵袭及迁移能力,这种改变可能与E-Cadherin、N-Cadherin、Vimentin的表达逆转EMT进程有一定相关性。Objective:To study the effect of long non-coding RNA(lncRNA)MEG3 on breast cancer cell invasion and migration and explore its mechanism.Methods:The expression of MEG3 in MDA-MB-231,MCF-7 and SKBR-3 in breast cancer cells was detected by real-time fluorescence quantitative PCR(qRT-PCR).MCF-7 cell line with lower expression level was selected for further studies.Overexpressed MEG3 was constructed into pcDNA3.1 plasmid that was transfected in MCF-7,and then the changes of miRNA-21 were examined by qRT-PCR.The migration of MCF-7 cells was detected by scratch test.The invasion of MCF-7 cells was detected by transwell assays.The expression levels of E-cadherin,N-cadherin and Vimentin associated with EMT were detected by western blot.Results:The expression of miRNA-21 in the MCF-7 cell lines with overexpressed MEG3,as well as the invasion and migration ability of the cells were significantly decreased,together with t.The expression level of E-cadherin protein in the cells was significantly increased,and the expression level of N-cadherin and Vimentin was significantly decreased,the differences were statisti⁃cally significant(P<0.05).Conclusion:LncRNA MEG3 has influence on the invasion and migration of breast cancer cells by down-regulating miRNA-21,which may be correlated with the reversal of EMT process regulated by E-cadherin,N-cadherin and Vimentin.
