摘 要:目的分析基质金属蛋白酶1(MMP1)基因在肺癌细胞与正常细胞中的表达差异及其预后意义。方法从Oncomine数据库中收集近20年关于非小细胞肺癌中MMP1表达水平的研究信息,统计比较MMP1在肺癌细胞组织与正常细胞组织中的表达差异;采用CCLE数据库分析MMP1基因在不同癌细胞系中的表达情况,同时应用Kaplan Meier Plotter在线数据库分析MMP1表达水平与非小细胞肺癌患者预后的相关性。结果共收集到MMP1在多种类型肿瘤中表达水平的研究338项,其中有统计学差异的研究结果 62项,包括MMP1表达水平升高61项,MMP1表达水平下降1项。进一步筛选分析发现,有8项为非小细胞肺癌的研究,均显示MMP1在肺癌细胞中呈高表达,与正常细胞比较差异显著(P<0.05)。CCLE数据库分析显示了MMP1在40类癌细胞系中的表达情况,其在非小细胞肺癌中的表达值排23位。Kaplan Meier Plotter统计分析显示,MMP1表达水平对肺腺癌患者预后有显著影响HR=1.53(1.21~1.94)(P=0.000 34),但对鳞状细胞肺癌患者预后无显著影响HR=0.91(0.72~1.15)(P=0.43)。结论 Oncomine、CCLE、Kaplan Meier Plotter数据库分析显示,MMP1在非小细胞肺癌中呈较高水平表达,且MMP1表达与肺腺癌患者预后明显相关,这为临床研究肿瘤相关基因提供了指导。Objective To study the expression difference of matrix metalloproteinase-1(MMP1) gene between lung cancer cells and normal cells and its prognostic significance.Methods The information about MMP1 expression in non-small cell lung cancer(NSCLC) in recent 20 years was collected from Oncomine database,and the difference of MMP1 expression between lung cancer cells and normal cells was compared.CCLE database was used to analyze the expression of MMP1 gene in different cancer cell lines.The Kaplan Meier Plotter online database was used to analyze the correlation between MMP1 expression and prognosis of patients with NSCLC.Results A total of 338 studies on the expression level of MMP1 in various types of tumors were collected,of which 62 were statistically different,including 61 with increased MMP1 expression level and 1 with decreased MMP1 expression level.Further screening analysis showed that 8 studies of NSCLC showed high expression of MMP1 in lung cancer cells,which was significantly different from normal cells(P<0.05).CCLE database analysis showed the expression of MMP1 in 40 cancer cell lines,and its expression value in non-small cell lung cancer ranked 23.Kaplan Meier Plotter analysis showed that the expression level of MMP1 had a significant impact on the prognosis of patients with lung adenocarcinoma,HR=1.53(1.21~1.94)(P=0.000 34),but had no significant impact on the prognosis of patients with squamous cell lung cancer,HR=0.91(0.72~1.15)(P=0.43).Conclusion Oncomine,CCLE and Kaplan Meier plotter database analysis show that MMP1 is highly expressed in NSCLC,and the expression of MMP1 is significantly correlated with the prognosis of patients with lung adenocarcinoma,which provides guidance for clinical study of tumor related genes.
