摘 要:目的探讨当归多糖对大鼠视网膜缺血-再灌注损伤的保护作用及机制。方法SD大鼠随机分为对照组、模型组、当归多糖低、高浓度组,每组10只。对照组不做特殊处理;模型组、当归多糖低、高浓度组建立视网膜缺血-再灌注损伤模型。取4组大鼠视网膜组织,观察其在HE染色及透射电镜下的细胞层面结构变化。检测并比较4组丙二醛(MDA)含量,超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性,核因子E2相关因子2(Nrf2)、血红素加氧酶-1(HO-1)蛋白含量。结果与对照组比较,当归多糖低、高浓度组视网膜组织损伤程度逐渐减轻。与对照组比较,模型组大鼠视网膜组织中MDA含量升高,SOD、GSH-Px活性降低,差异有统计学意义(P<0.05)。与模型组比较,当归多糖低、高浓度组大鼠视网膜组织中MDA含量下降,SOD、GSH-Px活性升高,且当归多糖高浓度组较低浓度组变化更显著,差异均有统计学意义(P<0.05)。与对照组比较,模型组大鼠视网膜组织中Nrf2、HO-1蛋白表达水平显著下降(P<0.05)。与模型组比较,当归多糖低、高浓度组视网膜组织Nrf2、HO-1蛋白表达显著升高,当归多糖高浓度组高于当归多糖低浓度组(P<0.05)。结论当归多糖可以通过激活Nrf2-抗氧化反应元件信号通路降低SD大鼠视网膜氧化应激反应,缓解缺血性视网膜损伤。Objective To investigate the protective effect of Angelica Sinensis Polysaccharide on rats with retinal ischemia-reperfusion injury and its mechanism.Methods SD Rats were randomly divided into normal control group,model group,low and high concentrations Angelica Sinensis Polysaccharide treatment groups(n=10 in each group).Control group was not given special treatment,while model,low and high concentrations Angelica Sinensis Polysaccharide treatment groups were established retinal ischemia-reperfusion injury models.Retinal tissues of the four groups were taken off to observe changes of cell level structure under HE staining and projection electron microscope.Contents of malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)activity,nuclear factor E2 related factor 2(Nrf2)and heme oxygenase-1(HO-1)protein were detected and compared among the four groups.Results Compared with that in control group,damage degrees of retina in low and high concentrations Angelica Sinensis Polysaccharide treatment groups were gradually reduced.Compared with those in control group,MDA content in retina was increased,while activities of SOD and GSH-Px were decreased in model group(P<0.05).Compared with those in model group,MDA contents in retina were decreased,while activities of SOD and GSH-Px were increased in low and high concentrations Angelica Sinensis Polysaccharide treatment groups,and the changes in high concentration Angelica Sinensis Polysaccharide treatment group were more significant than those in low concentration Angelica Sinensis Polysaccharide treatment group(P<0.05).Compared with those in control group,protein expressions of Nrf2 and HO-1 in model group were significantly decreased(P<0.05).Compared with those in model group,protein expressions of Nrf2 and HO-1 in treatment groups were significantly higher,and the expressions in high concentration treatment group were higher than those in low concentration treatment group(P<0.05).Conclusion Angelica Sinensis Polysaccharide may relieve ischemic retin
