摘 要:目的:研究RAB10在人肝癌组织中的表达及其与患者临床预后的关系。方法:Western Blot检测肝癌组织与正常肝组织中RAB10的表达情况,利用TCGA数据库分析RAB10的表达与肝癌患者临床病理特征的关系以及对预后的影响,进一步分析与RAB10高表达可能相关的ATG蛋白。结果:与正常肝脏组织相比,RAB10在肝癌组织中的表达显著升高;RAB10的表达情况与肿瘤TNM分期显著相关,表达越高,分期越差(P<0.01);Kaplan-Meier分析显示在肝癌组织中RAB10高表达的患者无病生存期(DFS)、总生存期(OS)均显著低于低表达的患者(P<0.001)。在肝癌组织中,RAB10与多种ATG的表达呈明显的正相关性。结论:RAB10在肝癌组织中的表达明显上调可能预示肝癌患者预后差。RAB10可作为肝癌预后检测的分子标志物及治疗的潜在靶点。RAB10的高表达与肝癌患者不良预后的关系可能通过与ATG家族成员发生相互作用有关。Objective:To investigate the expression of RAB10 in human liver cancer tissues and its relationship with the clinical prognosis of patients.Methods:Western blot was used to detect the expression of RAB10 in human liver cancer tissue and normal liver tissue.The TCGA database was used to analyze the relationship between RAB10 expression and the clinicopathological characteristics of liver cancer patients and the relationship with clinical prognosis.Further,we analyzed the correlation between ATG and RAB10 expression.Results:Compared with normal liver tissues,the expression of RAB10 in human liver cancer tissues was significantly increased;the expression of RAB10 was significantly related to the tumor TNM stage,and the higher the expression was,the worse the stage was(P<0.01);Kaplan Meier analysis showed that patients with high expression of RAB10 in liver cancer tissues had significantly lower disease-free survival(DFS)and overall survival(OS)than those with low expression(P<0.01).In liver cancer tissues,RAB10 was significantly positively correlated with the expression of various ATGs.Conclusion:The significant up-regulation of RAB10 expression in liver cancer tissues may indicate a poor prognosis for patients with liver cancer.RAB10 can be used as a molecular marker for prognostic detection of liver cancer and a potential target for treatment.The relationship between the high expression of RAB10 and the poor prognosis of patients with liver cancer may be related to the interaction with members of the ATG family.
