摘 要:目的建立高效液相色谱法(high performance liquid chromatography,HPLC)同时测定榅桲籽中蔷薇酸和委陵菜酸含量的分析方法。方法样品经石油醚脱脂后,用甲醇超声提取,采用Kromasil 100-5-C_(18)(4.6 mm×250 mm, 5μm)色谱柱分离,以甲醇-0.2%磷酸(70:30, V:V)作为流动相,流速1.0 mL/min,柱温30℃,进样体积为10μL,在210nm波长下进行检测。结果蔷薇酸和委陵菜酸分别在7.92~55.44μg/mL和3.76~18.8μg/mL浓度范围内线性关系良好,相关系数均大于0.999,加样回收率为90.4%~98.2%(n=6)。结论该方法简便、快速、准确、重现性好,可用于榅桲籽中蔷薇酸和委陵菜酸的快速测定。Objective To establish a method for the simultaneous determination of euscaphic acid and tormentic acid in Cydonia oblonga seeds by high performance liquid chromatography.Methods Sample were defatted by petroleum ether and extracted with methanol.The chromatographic separation was achieved on a Kromasil 100-5-C18 column(4.6 mm×250 mm,5μm)with methanol-0.2%phosphoric acid(70:30,V:V)as mobile phase,the flow rate was 1.0 mL/min,the column temperature was 30℃,injection volume was 10μL,and detection wavelength was 210 nm.Results The euscaphic acid and tormentic acid were separated well and showed good linearity in the range of 7.92‒55.44 g/mL and 3.76‒18.8 g/mL,and the correlation coefficients were all greater than 0.999.The average recoveries were 90.4%‒98.2%(n=6).Conclusion This method is simple,rapid,accurate,credible and repeatable.It can be used for determination of euscaphic acid and tormentic acid in Cydonia oblonga seeds.
