摘 要:目的基于阿尔兹海默症神经胶质细胞增生所致炎症反应的研究。方法2019年7月—2020年1月实验选取雄性小鼠16只,随机分为两组,各8只。模型组中双侧海马注射凝胶态Aβ致阿尔兹海默症,对照组中注射生理盐水,硫堇染色观察皮层神经元形态变化及数量,酶联免疫吸附试验观察小鼠血清中炎症因子变化,免疫组化检测胶质细胞、神经胶质原纤维酸性蛋白质表达。结果模型组大脑皮层神经元炎症数量(516.65±24.26)个,多于对照组(105.65±14.26)个,差异有统计学意义(t=41.310,P<0.05);模型组TNF-α、IL-1β分别为(85.65±25.56)pg/mL、(13.32±5.65)pg/mL,对照组分别为(40.05±13.12)pg/mL、(7.44±2.45)pg/mL,模型组小鼠血清中TNF-α和IL-1β水平高于对照组,差异有统计学意义(t=4.489、2.701,P<0.05)。结论阿尔兹海默病中小鼠神经元炎症细胞数量显著偏多,胶质细胞及促炎因子释放会造成小鼠脑神经元损伤。Objective To study the inflammatory response caused by the proliferation of glial cells in Alzheimer's disease.Methods From July 2019 to January 2020,16 male mice were selected for the experiment,and they were randomly divided into two groups with 8 mice each.In the model group,the bilateral hippocampus was injected with gelatinous Aβ to cause Alzheimer's disease,and the control group was injected with physiologysaline and thionine staining were used to observe the morphological changes and number of cortical neurons,enzyme-linked immunosorbent assay to observe the changes of inflammatory factors in the serum of mice,and immunohistochemistry to detect the expression of glial cells and glial fibrillary acidic proteins.Results The number of cerebral cortex neuron inflammation in the model group was(516.65±24.26)pcs,which was more than that of the control group(105.65±14.26)pcs,and the difference was statistically significant(t=41.310,P<0.05).TNF-α and IL-1β in the model group were(85.65±25.56)pg/mL,(13.32±5.65)pg/mL,the control group were(40.05±13.12)pg/mL,(7.44±2.45)pg/mL,the serum levels of TNF-α and IL-1β in the model group were higher than those in the control group,and the difference between the two groups was statistically significant(t=4.489,2.701,P<0.05).Conclusion The number of neuronal inflammatory cells in Alzheimer's disease is significantly higher,and the release of glial cells and pro-inflammatory factors can cause brain neuron damage in mice.
