摘  要:目的探讨AT丰富结合域1A(ARID1A)和真核细胞起始因子4E(eIF4E)在胃癌组织表达的临床意义。方法选取2015年3月至2020年7月广东医科大学惠州临床医学院(惠州市中心人民医院)和广东医科大学附属医院病理学确诊的94例胃癌组织以及对应癌旁组织,应用免疫组织化学方法检测上述组织中ARID1A蛋白以及eIF4E蛋白表达水平,采用χ^(2)检验分析ARID1A蛋白和eIF4E蛋白与胃癌临床病理特征之间的关系。结果ARID1A蛋白在胃癌组织表达率为32.98%(31/94)、对应癌旁组织ARID1A蛋白表达率为80.85%(76/94),两者差异有统计学意义(χ^(2)=43.925、P<0.01),ARID1A表达水平和胃癌组织学分化程度、TNM分期、淋巴结转移明显相关(χ^(2)=5.307、6.812、4.583,P<0.05)。eIF4E蛋白在胃癌组织表达率为79.79%(75/94)、对应癌旁组织eIF4E蛋白表达率为22.34%(21/94),两者差异有统计学意义(χ^(2)=62.071,P<0.01),eIF4E表达水平和胃癌TNM分期、淋巴结转移明显相关(χ^(2)=6.479、5.379,P<0.05)。结论ARID1A蛋白在胃癌组织中表达下调,eIF4E蛋白在胃癌组织中表达上调,ARID1A和eIF4E有助于胃癌临床诊断及评估患者预后。Objective To investigate the clinical significance of AT-rich interactive domain 1A(ARID1A)and eukaryotic initiation factor 4E(eIF4E)in gastric carcinoma.Methods The expression levels of ARID1A protein and eIF4E protein in 94 cases of gastric carcinoma tissues and adjacent gastric tissues of patients pathologically confirmed from March 2015 to July 2020 on Huizhou Clinical Medical College of Guangdong Medical University(Huizhou Municipal Central Hospital)and Affiliated Hospital of Guangdong Medical University were examined by envision immunohistochemical method.Chi-square test was used to analyze the relationship between the expression deletion of ARID1A and eIF4E protein and clinicopathological parameters of the patients.Results ARID1A expression rate in gastric carcinoma tissues and the adjacent gastric tissues was 32.98%(31/94)and 80.85%(76/94)(χ^(2)=43.925,P<0.01).The expression of ARID1A was related gastric carcinoma to histology grade,tumor node metastasis(TNM)stage,lymph node metastasis(respectively,χ^(2)=5.307,6.812,4.583,P<0.05).The eIF4E expression rate in gastric carcinoma tissues and the adjacent gastric tissues was 79.79%(75/94)and 22.34%(21/94)respectively(χ^(2)=62.071,P<0.01).The expression of eIF4E was related gastric carcinoma to TNM stage,lymph node metastasis(respectively,χ^(2)=6.479,5.379,P<0.05).Conclusion ARID1A protein expression is downregulated in gastric carcinoma tissues,and eIF4E protein expression is upregulated in gastric carcinoma tissues.The expression levels of ARID1A protein and eIF4E protein were useful markers for predicting the metastasis and prognosis of gastric carcinoma.
