摘　　要：目的观察长链非编码RNA(lncRNA)-linc00092在胃癌组织中的分布特点,探讨linc00092表达高低对胃癌患者多种生存特点的影响,并对其与胃癌常见临床病理学特点的相关性进行分析。方法在13例胃癌患者中,分析癌组织和正常组织中的linc00092的表达差异;在线生存数据库中,分析胃癌组织中的linc00092表达水平对胃癌患者总生存(OS)、疾病进展后生存(PPS)、首次进展后生存(FP)的影响;收集90例胃癌患者癌组织新鲜冰冻组织标本,行实时定量聚合酶链反应(Real-time PCR)检测linc00092水平,分析癌组织中的linc00092与胃癌患者常见临床病理特点的关系。采用χ^(2)检验或者Fisher′s确切概率法分析。结果13对胃癌组织和正常组织中,linc00092的表达水平在胃癌组织的表达水平是胃正常组织中的2.316倍(t=3.195,P<0.05),差异有统计学意义;linc00092高表达的胃癌人群的OS在数据库胃癌总体人群、Ⅲ~Ⅳ期胃癌患者、女性患者、男性患者、仅接受手术患者、人表皮生长因子受体2(Her-2)阴性和阳性患者中明显低于linc00092低表达者(P<0.05);linc00092高表达的胃癌人群的FP在总体人群、Ⅱ~Ⅲ期胃癌患者、Her-2阴性和阳性患者中显著低于linc00092低表达组(P<0.05),差异均有统计学意义;linc00092高表达的胃癌人群的PPS在总体人群、Ⅰ~Ⅳ期胃癌患者、女性和男性患者、仅接受手术患者、Her-2阴性和阳性患者中显著低于linc00092低表达组(P<0.05),差异均有统计学意义;胃癌中的linc00092的表达水平越高,癌组织分化越差(P<0.05),肿瘤越容易出现淋巴结转移(P<0.05)、更容易出现深部浸润(P<0.05),差异均有统计学意义。结论较高的linc00092提示胃癌患者预后较差,同时linc00092在胃癌组中显著高表达,和患者的组织学分级、浸润深度和淋巴结转移情况明显相关。Objective To evaluate the expression levels of linc00092 in gastric cancer and analyze the correlation between linc00092 and clinicopathological features,and effects of linc00092 on overall survival(OS),first progression(FP)and post-progression survival(PPS)in gastric cancer patients.Methods In 13 gastric cancer patients,the expression levels of linc00092 in gastric cancer samples and normal tissues were detected.Real-time quantitative polymerase chain reaction(Real-time PCR)was utilized to analyze levels of linc00092.The correlation between linc00092 and the clinicopathological features in gastric cancer was analyzed.Online Kaplan-Meier Plotter(KM)database was used to evaluate the effects of linc00092 on survival of gastric cancer patients.Results The linc00092 levels were higher in cancer samples than those in normal ones in gastric caner(fold change 2.316,t=3.195,P<0.01).Linc00092 levels were correlated to grade,tumor infiltration and lymph node metastasis positively(χ^(2)=4.712,8.478,7.608,P<0.05).OS rate of gastric cancer patients with higher levels of linc00092 were shorter than those with lower levels of linc00092 expression in all patients,stageⅢ-Ⅳ,female patients,male patients,surgery alone,human epidermal growth factor receptor 2(Her-2)negative and Her-2 positive groups(P<0.05).Similar trends were also observed in FP analysis in all patients,stageⅡ-Ⅲ,Her-2 negative and Her-2 positive groups(P<0.05),and in PPS analysis in all patients,stageⅠ-Ⅳ,female patients,male patients,surgery alone,Her-2 negative and Her-2 positive groups(P<0.05)Conclusion Linc00092 level was higher in gastric cancer tissues,and correlated to grade,tumor infiltration and lymph node stage positively.Higher expression of linc00092 is related to shorter survival in gastric cancer.
