摘 要:目的:探讨健脾化痰方对胃癌细胞PD-L1表达的影响,初步探讨其干预肿瘤免疫逃逸的机制。方法:40只大鼠随机分为生理盐水对照组及健脾化痰方高、中、低剂量组,每组10只,采用灌胃方法,制备不同浓度含药血清。各组含药血清处理胃癌细胞(SGC-7901)24 h。采用流式表面标记技术检测SGC-7901细胞上PD-L1的表达,采用免疫荧光染色法进行验证。结果:与对照组比较,健脾化痰方低、中、高浓度含药血清处理的胃癌细胞SGC-7901,24 h后PD-L1表达明显下降,平均荧光强度表达量明显降低,P<0.01,且程剂量依赖型,免疫荧光染色法可进一步验证其结果。结论:健脾化痰方含药血清能降低胃癌细胞SGC-7901中PD-L1表达,且程剂量依赖型,其可能通过这一机制干预肿瘤的免疫逃逸,起到抗肿瘤作用。Objective:To investigate the effect of Jianpi Huatan Formula on the expression of PD-L1,and to explore its intervention mechanism in immune escape.Methods:40 rats were randomly divided into the normal saline control group and the Jianpi Huatan Formula groups of low-dose,medium-dose and high-dose,with 10 rats in each group.Different concentrations of drug-containing serum were prepared to intervene gastric cancer cells SGC-7901 for 24 hours.Flow cytometry was used to analyze the expression of PD-L1 on SGC-7901 cells.Immunofluorescence staining was used for the verification.Results:Compared to that in the control group,the expression level of PD-L1 was significantly decreased in gastric cancer cells SGC-7901 in a dose-dependent manner 24 hours after drug-containing serum intervention(P<0.01),which was verified by immunofluorescence staining method.Conclusion:Drug-containing serum of Jianpi Huatan Formula can down-regulate the expression of PD-L1 in a dose-dependent manner in gastric cancer cells SGC-7901.It is possible to intervene the immune escape of tumor through this mechanism and play an anti-tumor role.
