摘 要:目的分析miR-140在结肠癌中的作用机制。方法qRT-PCR检测miR-140在结肠癌细胞和组织中的表达;CCK8法、细胞划痕实验检测miR-140对结肠癌HT29细胞增殖和迁移的影响。TargetScan筛选miR-140的潜在靶基因并通过双荧光素酶报告基因试验进行验证;采用qRT-PCR和Western blot检测miR-140对SOX2表达的影响;检测SOX2在结肠癌细胞和组织中的表达;裸鼠皮下成瘤实验研究miR-140和SOX2对肿瘤生长的影响。结果miR-140在结肠癌组织和细胞中表达降低(P<0.01)。过表达miR-140能明显抑制结肠癌细胞HT29的增殖和迁移。SOX2是miR-140靶基因,并在结肠癌组织和细胞株表达升高(P<0.01);体内实验发现miR-140过表达能抑制SOX2的表达,以及细胞增殖及肿瘤生长(P<0.05)。结论miR-140通过靶向调控SOX2,从而抑制结肠癌细胞的增殖和迁移及裸鼠移植瘤的生长,在结肠癌中起抑癌作用。Objective The purpose of this study was to analyze the mechanism of miR-140 in colon cancer.Methods The expression of miR-140 in colon cancer cells and tissues was detected by qRT-PCR,the effect of miR-140 on the proliferation and migration of colon cancer HT29 cells was detected by CCK8 assay and scratch-wound assay,TargetScan was used to screen potential target genes of miR-140 and was verified by double luciferase reporter assay,the effect of miR-140 on the expression of SOX2 was detected by qRT-PCR and Western blot,the expression of SOX2 in colon cancer cells and tissues was detected,tumorigenesis in nude mice was performed to investigate the effects of miR-140 and SOX2 on tumor growth.Results miR-140 expression was decreased in colon cancer tissues and cells(P<0.01)Overexpression of miR-140 significantly inhibited the proliferation and migration of colon cancer cell HT29,SOX2 was a miR-140 target gene,and its expression was increased in colon cancer tissues and cell lines(P<0.01)Overexpression of miR-140 was found to inhibit the expression of SOX2,cell proliferation and tumor growth in vivo(P<0.05).Conclusion miR-140 inhibits the proliferation and migration of colon cancer cells and the growth of transplanted tumors in nude mice through targeted regulation of SOX2,which plays a role in cancer suppression in colon cancer.
