摘　　要：目的探究胃肠型HIV卡波西肉瘤组织中人类8型疱疹病毒(HHV8)感染与ORF42、哺乳动物雷帕霉素靶蛋白复合物1(mTORC1)表达的关系。方法选取2015年9月至2019年9月新疆维吾尔自治区人民医院收治的艾滋病合并胃肠型卡波西肉瘤患者36例作为研究对象。留取入组患者卡波西肉瘤组织及瘤旁组织,采用免疫组织化学染色检测胃肠型HIV卡波西肉瘤组织及瘤旁组织中HHV8表达情况,Western blotting法检测ORF42、mTORC1蛋白表达水平,qRT-PCR法检测ORF42、mTORC1、血管内皮生长因子(VEGF)、白介素-6(IL-6)mRNA表达水平,Pearson法分析胃肠型HIV卡波西肉瘤组织中ORF42、mTORC1与VEGF、IL-6相关性。结果胃肠型HIV卡波西肉瘤组织中HHV8阳性率明显高于瘤旁组织(P<0.05)。胃肠型HIV卡波西肉瘤组织中ORF42、mTORC1蛋白及mRNA,VEGF、IL-6 mRNA表达水平明显高于瘤旁组织(P<0.05)。感染亚组卡波西肉瘤组织中ORF42、mTORC1蛋白及mRNA,VEGF、IL-6 mRNA表达水平明显高于未感染亚组(P<0.05)。胃肠型HIV卡波西肉瘤组织中ORF42、mTORC1 mRNA与VEGF、IL-6 mRNA表达水平均呈正相关(P<0.05)。结论胃肠型HIV卡波西肉瘤组织中HHV8感染率增加,可能通过激活ORF42、mTORC1表达促进肿瘤发生发展。Objective To investigate the relationships between human herpesvirus 8(HHV8)infection and the expressions of ORF42 and mammalian target of rapamycin complex 1(mTORC1)in gastrointestinal HIV Kaposi’s sarcoma tissues.Methods From Sep.2015 to Sep.2019,36 patients with gastrointestinal HIV Kaposi’s sarcoma were selected as the study objects.Kaposi’s sarcoma and its peritumoral tissues were collected,the expression of HHV8 in gastrointestinal HIV Kaposi’s sarcoma tissues and its peritumoral tissues was detected by immunohistochemistry,Western blotting was used to detect the protein expressions of ORF42 and mTORC1,the mRNA levels of ORF42,mTORC1,vascular endothelial growth factor(VEGF)and interleukin-6(IL-6)were detected by qRT-PCR,and Pearson’s method was used to analyze the correlations between ORF42,mTORC1 and VEGF,IL-6 in gastrointestinal HIV Kaposi’s sarcoma.Results The positive rate of HHV8 in gastrointestinal HIV Kaposi’s sarcoma tissues was significantly higher than that in peritumoral tissues(P<0.05).The expressions of ORF42,mTORC1 proteins and mRNAs,VEGF,IL-6 mRNAs in gastrointestinal HIV Kaposi’s sarcoma tissues were significantly higher than those in adjacent tissues(P<0.05).The expressions of ORF42,mTORC1 proteins and mRNAs,VEGF,IL-6 mRNAs in Kaposi’s sarcoma tissues of the infected subgroup were significantly higher than those in uninfected subgroup(P<0.05).The expression levels of ORF42 and mTORC1 mRNAs were positively correlated with VEGF and IL-6 mRNAs(P<0.05).Conclusion The infection rate of HHV8 is increased in gastrointestinal HIV Kaposi’s sarcoma,which may promote tumor development by activating the expressions of ORF42 and mTORC1.
