高效液相色谱法快速测定红糖中丙烯酰胺

摘　　要：建立了红糖良好水溶性样品的高效液相色谱法检测丙烯酰胺含量的方法。样品未经前处理,直接溶解进样,对色谱柱的选择、紫外检测波长、流动相中有机相比例等实验条件进行优化和考察。结果表明:采用Hypercarb多孔石墨碳液相色谱柱(4.6 mm×100 mm,3μm),在流动相为甲醇-去离子水(15︰85,V/V),流速为1.0 mL/min,紫外检测波长为210 nm的条件下,红糖中丙烯酰胺得到了良好的分离。该方法在0.01~0.16μg/mL范围内线性良好,相关系数为0.9991。丙烯酰胺的检出限为0.003μg/mL,定量限为0.01μg/mL,平均回收率为86.4%~93.4%,日内精密度在1.06%~3.29%,日间精密度在2.34%~4.40%。该方法能有效检测出红糖中的丙烯酰胺,前处理简单,灵敏度高,适用于红糖中丙烯酰胺的日常检测。An efficient high performance liquid chromatography(HPLC)method was developed for the determination of acrylamide in brown sugar.The selection of chromatographic columns,the UV detection wavelength,the mobile phase composition and other experimental conditions were investigated and optimized,respectively.The mobile phase was composed of a mixture of methanol and deionized water(15:85,V/V)at a flow rate of 1.0 mL/min.Under the optimal conditions(UV detection wavelength 210 nm),the analyte was well separated.The linear range was 0.01~0.16μg/mL with a correlation of 0.9991.The limit of detection(LOD)of acrylamide was 0.003μg/mL,the limit of quantitative(LOQ)was 0.01μg/mL.The average recoveries were at the range of 86.4%~93.4%,The intra-day precision ranged from 1.06%to 3.29%,and the inter-day precision ranged from 2.34%to 4.40%.The method has been used for the determination of acrylamide in brown sugar with good results.
