Hp阳性胃癌组织miR-1290、miR-490-3p表达变化及其临床意义

摘　　要：目的探讨幽门螺旋杆菌(Hp)阳性胃癌组织微小RNA-1290(miR-1290)、微小RNA-490-3p(miR-490-3p)表达变化及其临床意义。方法选择胃癌患者92例,其中Hp阳性61例、Hp阴性31例。采用RT-qPCR法检测胃癌组织及其配对的癌旁正常组织miR-1290、miR-490-3p表达。分析胃癌组织miR-1290表达与miR-490-3p表达的关系,以及Hp阳性胃癌组织miR-1290、miR-490-3p表达与患者临床病理特征的关系。分别以HP阳性胃癌组织miR-1290、miR-490-3p相对表达量的中位数为临界值,将患者分为miR-1290低表达者与miR-1290高表达者、miR-490-3p低表达者与miR-490-3p高表达者,比较不同miR-1290、miR-490-3p表达患者5年总生存率。结果与癌旁正常组织比较,Hp阳性与阴性胃癌组织miR-1290表达上调,miR-490-3p表达下调(P均<0.05)。无论是Hp阳性胃癌组织还是Hp阴性胃癌组织,miR-1290表达与miR-490-3p表达均无明显相关性(P均>0.05)。miR-1290、miR-490-3p表达均与Hp阳性胃癌淋巴结转移、远处转移和TNM分期有关(P均<0.05)。miR-1290低表达者30例、miR-1290高表达者31例,miR-490-3p低表达者30例、miR-490-3p高表达者31例。miR-1290低表达者5年总生存率明显高于miR-1290高表达者,miR-490-3p低表达者5年总生存率明显低于miR-490-3p高表达者(P均<0.05)。结论Hp阳性胃癌组织miR-1290表达上调、miR-490-3p表达下调,其表达变化与肿瘤淋巴结转移、远处转移、TNM分期和患者预后有关。Objective To investigate the expression and clinical significance of microRNA-1290(miR-1290)and microRNA-490-3p(miR-490-3p)in the Hp positive gastric cancer.Methods Ninety-two patients with gastric cancer were selected,in which 61 cases were Hp positive and 31 cases were Hp negative.The expression of miR-1290 and miR-490-3p in the gastric cancer tissues and adjacent tissues were detected by RT-qPCR.The correlation between the expres⁃sion of miR-1290 and miR-490-3p in gastric cancer was analyzed,and the relationship between the expression of miR-1290 and miR-490-3p in gastric cancer tissues,and the relationship of miR-1290 and miR-490-3p expression with the clin⁃icopathological characteristics of Hp positive gastric cancer patients were analyzed.The median relative expression of miR-1290 and miR-490-3p in Hp positive gastric cancer tissues were taken as the critical value,and the patients were then di⁃vided into patients with low expression of miR-1290 and patients with high expression of miR-1290,patients with low ex⁃pression of miR-490-3p,and patients with high expression of miR-490-3p,respectively.The 5-year overall survival rate of patients with different expression of miR-1290 and miR-490-3p was compared.Results Compared with the adjacent tis⁃sues,the expression of miR-1290 was up-regulated in Hp positive and Hp negative gastric cancer tissues,while the expres⁃sion of miR-490-3p was down-regulated(both P<0.05).Whether in Hp positive or Hp negative gastric cancer tissues,there was no correlation between the expression of miR-1290 and miR-490-3p in the gastric cancer tissues(both P>0.05).The expression of miR-1290 and miR-490-3p were related to distal metastasis,lymph node metastasis and TNM stage of Hp positive gastric cancer(all P<0.05).There were 30 cases of low expression of miR-1290 and 31 cases of high expres⁃sion of miR-1290,30 cases of low expression of miR-490-3p,and 31 cases of high expression of miR-490-3p.The 5-year overall survival rate of patients with low expression of miR-1290 was
